摘 要:目的:研究模拟失重模型动物前扣带回皮质(anterior cingulated cortex,ACC)神经元激活状况及其变化的时间模式。方法:将SD大鼠随机分成4组。所有动物采用吊尾、头低位方式使后肢去负荷(hind-limb unloading,HU)方式模拟失重生理。第一、二组分别经HU处理1 d和4 d。第三、四组动物分别经过HU 1 d和4 d,并进行双轴旋转运动刺激2 h。然后,动物灌流固定、取脑、切片(厚度25μm),采用常规免疫组织化学染色技术卵白素-生物素复合物(ABC)法对ACC内神经元Fos蛋白进行免疫反应,3,3′-二氨基联苯胺四盐酸盐水合物(DAB)为呈色剂。统计分析Fos阳性神经元数量变化。结果:(1)1-d HU组、4-d HU和1-d HU-旋转组和4-d HU-旋转组动物ACC内Fos阳性神经元数目分别是344.98±90.620、389.111±133.774、486.47±182.152和464.444±286.881。(2)HU处理1 d到4 d ACC神经激活数目有增加趋势,旋转刺激后ACC内神经元也有被激活趋势,但统计分析显示各组间无统计学差异。结论:HU处理使ACC内神经元有被激活趋势,但随HU处理时间的延长,ACC神经元活动对HU刺激产生适应。Objective:To investigate the neuronal activity in anterior cingulate cortex(ACC)and its changing pattern over time in rat model of weiglessness.Methods:Sprague-Dawley rats were divided randomly into 4 groups.All the animals were treated for simulation of weightlessness by way of tail-suspending with hind-limb unloaded(HU)and head downward.The first two groups received HU for 1 and 4 days respectively;the last two groups experienced 1-and 4-d HU followed by double-axis rotation for 2 hours.Animals were then perfused followed by brain removal and sectioning(25μm thick/section).Routine immunohistochemistry of avidin-biotin complex(ABC)method was used to perform Fos protein immunoreactivity with ACC neurons and 3,3′-diaminobenzidine tetrahydrochloride(DAB)as the chromogen.Finally,statistical analysis of Fos-positive neuron counts was conducted.Results:(1)The number of Fos immunoreactive neurons in 1-d HU,4-d HU,1-d HU/rotation and 4-d HU/rotation groups were 344.98±90.620,389.111±133.774,486.47±182.152 and 464.444±286.881,respectively.(2)During 1-d HU to 4-d HU,then number of activated ACC neurons demonstrated a tendency to increase;and rotary stimulation tended to increase the number of activated ACC neurons.However,no statistical significance was detected between the four groups.Conclusion:Treatment with HU tended to activate the ACC neurons;and with prolonged HU possibly made ACC neurons adapted to the treatment.
