摘  要:[目的]研究香砂平胃颗粒对正常小鼠及胃肠动力障碍模型小鼠胃排空、肠推进和血清中胃动素(MOT)、胃泌素(GAS)、生长抑素(SS)、P物质(SP)和生长激素释放肽(Ghrelin)含量的影响。[方法]灌胃给予正常小鼠及阿托品所致胃肠动力障碍模型小鼠香砂平胃颗粒高、中、低剂量(2.6g/kg、1.3g/kg、0.65g/kg),采用称重法计算小鼠胃残留率和小肠炭末推进法计算小鼠肠推进率,进一步应用ELISA方法检测小鼠血清中MOT、GAS、SS、SP及Ghrelin的含量,并利用RT-PCR技术检测下丘脑中GhrelinmRNA与生长激素促分泌素受体(GHSR)mRNA转录水平。[结果]在胃肠运动方面,香砂平胃颗粒高剂量组能显著促进正常小鼠的胃排空(P<0.05),并能拮抗阿托品所致小鼠的胃排空抑制作用(P<0.05),香砂平胃颗粒高、中、低剂量组均能显著拮抗胃肠动力障碍模型小鼠的小肠推进抑制作用(P<0.05或P<0.01);在胃肠激素方面,香砂平胃颗粒高剂量组能提高正常小鼠及胃肠动力障碍模型小鼠血清中MOT和GAS水平(P<0.05或P<0.01),且能上调胃肠动力障碍模型小鼠血清中Ghrelin含量与下丘脑中GhrelinmRNA转录水平(P<0.01)。[结论]香砂平胃颗粒能提高正常小鼠与胃肠动力障碍模型小鼠的胃肠动力,其作用机制可能与增加血清中MOT、GAS与Ghrelin含量有关。[Objective]To study the effects of Xiangsha Pingwei granule on gastric emptying,intestinal propulsion and motilin(MOT),gastrin(GAS),somatostatin(SS),substance-P(SP)and Ghrelin in serum of normal mice and gas⁃trointestinal motility disorder model mice[.Methods]Normal mice and atropine-induced gastrointestinal motility disor⁃der model mice were given high,medium and low doses of Xiangsha Pingwei granule(2.6 g/kg,1.3 g/kg,0.65 g/kg).The gastric residue rate of mice was calculated by weighing method and the intestinal propulsion rate of mice was calcu⁃lated by small intestinal carbon powder propulsion method.The contents of MOT,GAS,SS,SP and Ghrelin in mouse se⁃rum were detected by ELISA.At the same time,the mRNA transcription levels of Ghrelin and GHSR in hypothalamus were detected by RT-PCR.[Results]In gastrointestinal motility,high dose of Xiangsha Pingwei granule can signifi⁃cantly promote gastric emptying in normal mice(P<0.05),and can antagonize the inhibition of gastric emptying in atro⁃pine model mice(P<0.05).High,medium and low doses of Xiangsha Pingwei granule could significantly antagonize the small intestine inhibition of atropine model mice(P<0.05 or P<0.01).In the aspect of gastrointestinal hormone,high dose of Xiangsha Pingwei granule can improve the level of motilin and gastrin in the serum of normal mice and at⁃ropine model mice(P<0.05 or P<0.01).It can up regulate the content of Ghrelin in serum and the transcription level of Ghrelin mRNA in hypothalamus of atropine model mice(P<0.01).[Conclusion]Xiangsha Pingwei granule can im⁃prove gastrointestinal motility in normal mice and gastrointestinal motility disorder model mice.Its mechanism may be based on the increase of MOT,GAS and Ghrelin in serum.
