芒果苷作用于COX-2和5-LOX双靶点改善良性前列腺增生炎症

摘  要:目的研究芒果苷对良性前列腺增生(benign prostatic hyperplasia,BPH)炎症的治疗作用及其潜在的作用机制。方法Western Blot和q-PCR技术检测大鼠前列腺组织中环氧合酶-2(cyclooxygenase-2,COX-2)、5-脂氧合酶(5-lipoxygenase,5-LOX)在蛋白水平上和mRNA水平上的表达,并用计算机分子对接技术加以佐证。此外,对前列腺湿质量(prostate weight,PW)、前列腺指数(prostate index,PI)、前列腺组织学形态进行考察。结果Western Blot和q-PCR结果表明,芒果苷可抑制大鼠前列腺组织中COX-2和5-LOX在蛋白水平和mRNA水平上的表达,分子对接技术结果表明芒果苷与COX-2和5-LOX受体模型均有结合能力。同时,PW、PI及组织病理学切片结果均表明芒果苷可明显改善BPH炎症。结论芒果苷同时作用于COX-2和5-LOX双靶点,通过下调COX-2和5-LOX的表达,从而改善BPH炎症。Objective To investigate the therapeutic effects of mangiferin on inflammation of benign prostatic hyperplasia(BPH)and its potential mechanisms.Methods Western Blot was used to detect the expressions of COX-2 and 5-LOX in the prostate at the protein level,and q-PCR was used to detect at the mRNA level.The computer molecular docking was used to verify the results.Meanwhile,prostate weight(PW),prostate index(PI),and histological morphology of the prostate were also investigated.Results Western Blot and q-PCR results showed that mangiferin could inhibit the expressions of COX-2 and 5-LOX in the prostate at the protein level and the mRNA level.The results of computer molecular docking showed that mangiferin could bind to both COX-2 and 5-LOX receptor models.At the same time,the results of PW,PI and histopathology also revealed that mangiferin significantly ameliorated inflammation of BPH.Conclusion Mangiferin simultaneously acts on the dual targets of COX-2 and 5-LOX and ameliorates inflammation of BPH by down-regulating the expressions of COX-2 and 5-LOX.
