摘 要:【背景和目的】NAC转录因子是一类植物特有的转录因子,在植物生长发育、生物/非生物胁迫中发挥着十分重要的调控作用,在普通烟草中克隆NtNAC042基因。【方法】进行生物信息学分析、亚细胞定位、转录激活试验和表达模式分析,以及基因功能分析。【结果】(1)生物信息学分析表明,NtNAC042基因编码蛋白具有高度保守的NAC结构域;(2)亚细胞定位和转录激活试验结果表明,NtNAC042定位于细胞核中,具有转录激活活性;(3)表达模式分析表明,NtNAC042基因在根和衰老叶片中表达量较高且受干旱胁迫、盐胁迫、H2O2处理诱导;(4)NtNAC042过表达烟草植株抗盐性显著提高。【结论】NtNAC042作为NAC型转录因子,在烟草衰老落黄及生物和非生物逆境应答中发挥着重要的调控作用。[Background and Object] NAC transcription factors are a class of plant-specific transcription factors,which play important roles in plant growth and development,biotic/abiotic stress regulation,and cloning of NtNAC042 gene in Nicotiana tabacum.[Methods]To explore the biological function of the NtNAC042 gene in Nicotiana tabacum,the NtNAC042 gene was cloned from tobacco.Subsequently,bioinformatic analysis,expression profiles,and subcellular localization and transactivation activity assay of NtNAC042 were performed.Moreover,the NtNAC042-overexpressing plants were assessed.[Results] 1) The bioinformatic analysis revealed that NtNAC042 was proved to contain a typical NAC domain.2) The subcellular localization and transactivation activity assay indicated that NtNAC042 protein,which was localized in the nucleus,had transactivation activity.3) In addition,the results of the qRT-PCR analysis suggested that the NtNAC042 gene was highly expressed in the senescing leaves and significantly induced by multiple stress treatments,including drought,salt stresses,and H2O2.4) Furthermore,NtNAC042-overexpressing tobacco plants showed significantly higher resistance to salt stresses.[Conclusion] Therefore,NtNAC042 as a typical NAC transcription factor,may play an important regulatory role in tobacco aging and yellowing and biotic/abiotic stress response.
