摘 要:为了探讨黄秦艽水提物(WVBF)对ANIT诱导的小鼠胆汁淤积性肝损伤保护作用及机制研究,将雄性C57BL/6小鼠50只随机分为5组,即正常组(NC)、模型组(MC)、阳性对照熊去氧胆酸组(UDCA 45 mg/kg)、WVBF低剂量组(WVBF 25 mg/kg)和WVBF高剂量组(WVBF 50 mg/kg).灌胃给予WVBF,连续10 d,在第7天用ANIT油溶液(75 mg/kg)一次性灌胃造模,采集血液和肝脏样品测定血生化、肝脏病理以及FXR/SHP通路相关基因水平.结果显示:与模型组相比,WVBF给药组小鼠肝脏坏死情况明显好转,炎性细胞浸润减少,血清中ALT、AST、TBA、T-Bil-V和D-Bil-V明显下降,肝脏中Fxr、Shp、Mpr2和Bsep mRNA的水平显著上调,WVBF(50 mg/kg)组Ntcp的mRNA水平显著下调(P<0.05).这表明黄秦艽对ANIT诱导的小鼠胆汁淤积性肝损伤具有保护作用,其机制可能与调节胆汁酸的合成、分泌、转运和重吸收有关.To investigate the protective effect and mechanism of water extract of Veratrilla baillonill Franch(WVBF)on mouse cholestatic liver injury induced by ANIT,a total of 50 male C57BL/6 mice were randomly divided into 5 groups according to body weight:normal group(NC),model group(MC),positive control group treated with ursodeoxycholic acid(UDCA 45 mg/kg),low dose of WVBF group(WVBF 25 mg/kg)and high dose of WVBF group(WVBF 50 mg/kg).WVBF was given by gavage once a day for 10 days.On the seventh day,the model was made by gavage with ANIT oil solution(75 mg/kg).Blood and liver samples were collected to detect the serum biochemistry,liver pathology and FXR/SHP pathway related genes.The results showed that compared with the model group,the liver necrosis,inflammatory cell infiltration,ALT,AST,TBA,T-BIL-V and D-BIL-V in serum significantly decreased,the mRNA levels of Fxr,Shp,Mpr2 and Bsep in liver were significantly increased,and the mRNA level of Ntcp in WVBF(50 mg/kg)group significantly decreased(P<0.05).The results suggest that Veratrilla baillonill Franch has a protective effect on ANIT induced cholestatic liver injury in mice,and its mechanism may relate to the regulation of bile acid synthesis,secretion,transport and re-absorption.
