摘 要:【目的】为了更加全面地探究CBF基因家族在豌豆中的进化及生物学特征。【方法】通过生物信息学方法从基因家族鉴定、结构分析、蛋白基本理化性质、亚细胞定位、染色体定位、顺式作用元件、蛋白质相互作用与表达分析等方面,对豌豆PsCBF基因家族进行系统鉴定和分子特征分析。【结果】在豌豆中共鉴定出21个PsCBF基因,染色体定位显示18个PsCBF基因分布在1、2、4、6与7号染色体上,其余3个未组装到染色体上(PsCBF19~PsCBF21)。21个PsCBF基因编码蛋白的氨基酸数在123(PsCBF09)~349(PsCBF11),且均属于疏水性蛋白。亚细胞定位预测显示18个PsCBF基因(85.71%)定位在细胞核上,二级结构主要以无规则延伸为主。蛋白基序显示同一类群的基因都具相似的保守基序特征且该家族成员包含1~7个保守基序;同一类群的基因也具有相似的基因结构,且该家族基因含1~2个外显子。PsCBF基因启动子区分析表明其含有多种顺势作用元件,在激素反应,组织特异表达以及逆境响应中起作用。蛋白聚类网络图显示其蛋白可能在豌豆应对非生物胁迫中起调节作用。此外,本研究发现低温胁迫后K2SiO3处理下大部分PsCBF基因的表达发生显著变化。【结论】本研究结果将为进一步分析豌豆PsCBF转录因子调控碗豆生物学过程及功能、碗豆抗寒的分子鉴定以及抗寒资源创新利用研究提供参考依据。【Objective】The study was conducted to explore the evolutionary and biological characteristics of CBF gene family in pea.【Method】bioinformatics methods were used to systematically and molecularly identify the pea PsCBF gene family in terms of gene family identification,structural analysis,physical and chemical properties of protein,subcellular location,chromosome location,cis-acting elements,and protein interactions and expression.【Result】Twenty-one PsCBF genes were totally identified in pea,and it was showed that 18 PsCBF genes were distributed on chromosomes 1,2,4,6 and 7 through chromosomal location,and the other three genes did not equipped on chromosomes.The 21 PsCBF genes encoded proteins with amino acid numbers ranging from 123(PsCBF09)to 349(PsCBF11),which all belonged to hydrophobic proteins.The prediction of subcellular location showed that 18 PsCBF genes(85.71%)were located on the nucleus,and the secondary structure mainly exhibited random extension.Protein motifs showed that the genes in the same group had similar characteristics of conservative motif,and the family members contained 1-7 conserved motifs;and also they had similar gene structure with 1-2 exons.By the promoterregion analysis of PsCBF gene,it was showed that the promoter contained a variety of cis-acting elements,which played a role in hormone response,tissue-specific expression and stress response.The protein clustering network diagram showed that its protein might play a regulatory role in pea's response to abiotic stress.In addition,it was found that the expression of most PsCBF genes changed significantly under K2SiO3 stress.【Conclusion】The results of this study will provide a reference for further analysis of the mechanism and biological function of PsCBF in regulating the biological processes of pea,the molecular identification of cold resistance of pea,and the research on the innovative utilization of cold resistance resources.
