向天果中柠檬苦素类化合物对氧化损伤下INS-1细胞保护作用研究

摘  要:为了探究向天果中swietenine(Stn)及swietenolide(Std)对氧化损伤下INS-1细胞保护作用。使用不同浓度的过氧化氢作用于INS-1细胞4 h,CCK-8法筛选出合适的造模条件;将向天果正丁醇层活性成分Stn、Std和阳性药格列齐特(Gli)作用于INS-1细胞24 h,CCK-8法筛选出化合物的安全作用浓度;在过氧化氢损伤模型下,将化合物作用于INS-1细胞24 h后,分别检测细胞活力并计算细胞存活率和ROS水平变化;使用胰岛素ELISA试剂盒检测葡萄糖浓度分别为3.3 mM和16.7 mM刺激下的胰岛素分泌量,模拟BIS和GSIS下胰岛素分泌量。化合物Stn、Std可以提高氧化损伤下INS-1细胞的存活率,促进胰岛素分泌,降低ROS水平,促进细胞增殖,改善细胞凋亡状况。化合物Stn、Std可保护氧化损伤下INS-1细胞。In order to explore the protective effect of Swietenine(Stn)and Swietenolide(Std)on INS-1 cells under oxidative damage,INS-1 cells were treated with different concentrations of hydrogen peroxide for 4 h,and the appropriate modeling conditions were screened by CCK-8 method.Ins-1 cells were treated with Stn,Std and gliclazide(Gli)for 24 h.The safe concentration of the compounds was screened by CCK-8 method.Under the model of hydrogen peroxide injury,INS-1 cells were treated with the compound for 24 h,and the cell viability,cell survival rate and ROS level were detected,respectively.Insulin secretion stimulated by 3.3 mM glucose and 16.7 mM glucose was detected by insulin ELISA kit to simulate the insulin secretion under BIS and GSIS.The final results showed that compounds Stn and Std could improve the survival rate of INS-1 cells under oxidative injury,promote insulin secretion,reduce ROS level,promote cell proliferation,and improve cell apoptosis.Compounds Stn and Std could protect INS-1 cells from oxidative damage.
