摘 要:SWI/SNF家族是ATP依赖的染色质重塑复合物,对基因的表达调控具有重要作用.为了探究水稻中染色质重塑因子OsSWIB1的功能,对OsSWIB1基因表达模式进行分析.结果表明:OsSWIB1在水稻的叶、叶鞘及穗中表达量较高,而在茎、茎顶端分生组织及根中表达量较低;在水稻不同的生长发育时期,除在苗期表达量较低外,OsSWIB1在分蘖期、孕穗期、抽穗期及灌浆期均有较高表达.此外,OsSWIB1在短日照条件下的表达明显高于在长日照条件下的表达.利用CRISPR-Cas9技术对OsSWIB1基因进行靶向编辑,共获得20株T0代转基因植株,通过测序分析,鉴定出3个纯合编辑的突变体.对1个杂合编辑突变体的后代进行分离,获得了1个缺失200 bp的纯合突变体.The SWI/SNF family is an ATP-dependent chromatin remodeling complex,which plays an important role in gene expression regulation.To investigate the function of chromatin remodeling factor OsSWIB1 in rice(Oryza sativa),the expression pattern of OsSWIB1 gene was analyzed.The results showed that OsSWIB1 displayed higher expression levels in leaves,leaf sheathes and panicles,but lower expression levels in stems,shoot apical meristem and roots.Among the different growth and development stages of rice,OsSWIB1 displayed higher expression levels at tillering,booting,heading,and grain filling stages except for the seedling stage.Moreover,the expression level of OsSWIB1 under short day condition was significantly higher than that under long day condition.To reveal the biological function of OsSWIB1 gene,the osswib1 mutants was created using CRISPR-Cas9 gene editing technology.A total of 20 T0 transgenic plants were obtained and three homozygous osswib1 were identified by sequencing analysis.In addition,a homozygous mutant with 200 bp deletion was obtained from heterozygous editing mutant in progeny.
