摘  要:本研究采用F1代群体简化基因组测序(RAD-seq)技术,构建了光皮树亲本的遗传图谱。统计结果显示,SNP基因分型共57089个,校正后为57071个,基因型为hk×hk、nn×np和lm×ll。过滤与筛选后统计占比,hk×hk为28.59%,nn×np为14.52%,lm×ll为12.92%。通过对F1代进行连锁遗传图谱标记,9个连锁群综合遗传距离从93.26到241.02 cM不等,综合覆盖图距为1260.94 cM,平均每个连锁群1091个标记,标记数最大的连锁群是Lg1,标记数为1744个;标记数最小的连锁群是Lg9,只有579个。通过对SNP标记数、标记距离和覆盖图距3个参数指标进行相关性分析,SNP标记数与覆盖图距呈极显著正相关,相关系数0.907。本研究构建了光皮树高密度遗传图谱,为其农艺性状定位与基因挖掘奠定了良好的基础。In this research,the genetic map of the parents of Swida wilsoniana was constructed by using the F1 population simplified genome sequencing(RAD-seq)technology.A total of 57089 SNP genotypes were found,and the corrected geno types were 57071.After filtering the corrected data,hk×hk accounted for 28.59%,nn×np accounted for 14.52%,and lm×ll accounted for 12.92%.Furthermore,F1 populations were labeled with linked genetic map.The comprehensive genetic distance of 9 linked groups ranged from 93.26 cM to 241.02 cM,and the comprehensive coverage map distance was 1260.94 cM,with an average of 1091 markers per linked group.From which,Lg 1 had the largest number of markers,with 1744 markers,while Lg 9 had the smallest number with 579.The results of the correlation analysis among the number of SNP markers,the distance of SNP markers,and cover map distance,showed that there was a significant positive correlation between SNP markers and cover map distance,with a correlation coefficient of 0.907.This study have laid a good foundation for further investigation on the location of agronomic traits and gene mining of S.wilsoniana.
