摘 要:【目的】全基因组范围内鉴定白花泡桐(Paulownia fortunei)基因组中的碱性亮氨酸拉链转录因子(basic leucine zipper,bZIP)家族成员,并研究该家族基因在泡桐丛枝病发生过程中的作用。【方法】基于白花泡桐基因组图谱,利用隐马尔科夫模型、拟南芥bZIP蛋白质序列和本地BlastP程序,以E-value<0.001为阈值进行PfbZIP家族成员鉴定;使用MAGE7.0软件进行蛋白质序列比对及系统发育进化树构建,使用Tbtools软件进行基因结构及启动子顺式作用元件分析,并利用泡桐丛枝病发生前后及恢复过程中的转录组数据筛选与泡桐丛枝病发生相关的基因。【结果】鉴定到PfbZIP基因家族共有89个成员,根据系统进化分析将该家族基因分为13个亚家族;共线性分析发现该家族内发生了63次片段复制事件;顺式作用元件分析表明,PfbZIP基因家族可能响应光照、植物激素调控、生物与非生物胁迫等生物学过程。对泡桐丛枝病发生前后及恢复过程中的转录组数据分析发现,30个PfbZIP基因在发病前后显著差异表达,其中PfbZIP46基因表达在恢复过程中也有明显变化,故推测PfbZIP46可能与丛枝植原体入侵有关。【结论】在白花泡桐基因组中共鉴定出89个PfbZIP基因,片段复制事件可能是PfbZIP转录因子家族扩张的重要原因,PfbZIP46可能在丛枝植原体入侵泡桐过程中扮演重要角色。【Objective】The purpose of our study was to identify the basic leucine zipper(bZIP) gene family in the genome of Paulownia fortunei,and to study the role of this gene family member in the occurrence of Paulownia witches' broom(PaWB).【Method】Based on the genome map of P.fortunei,the PfbZIP gene family members were identified with E-value0.001 as the threshold by using hidden Markov model,Arabidopsis bZIP protein sequences and BlastP program;the MAGE7.0 software was used to perform protein sequence alignment and phylogenetic tree construction,and Tbtools software was used to analyze gene structure and promoter cis-acting elements.The PfbZIP genes related to PaWB occurrence were screened out through the comparison of transcriptome data before and after pathogenetic process as well as the recovery process of PaWB.【Result】A total of 89 PfbZIP gene family members were identified,which were divided into 13 subfamilies according to phylogenetic analysis;the collinearity analysis revealed that 63 fragment duplication events occurred in this family;cis-element analysis showed that the PfbZIP genes family might have a response to biological processes such as light,plant hormone regulation,biotic and abiotic stresses.Comparative analysis of the RNA-seq data before and after pathogenetic process as well as the recovery process of PaWB revealed that 30 PfbZIP genes,including PfbZIP46,were significantly differentially expressed before and after the occurrence of PaWB.Furthermore,the expression level of PfbZIP46 gene also continuously changed during the recovery process of PaWB-infected seedlings.Therefore,it was speculated that PfbZIP46 might be related to the process of phytoplasma infection.【Conclusion】A total of 89 PfbZIP genes in the genome of P.fortune was identified.The fragment duplication event may be a significant reason for the expansion of the PfbZIP transcription factor family,and PfbZIP46 may play an important role in the process of phytoplasma infection.
