摘 要:目的 通过响应面法优化传统制剂妇科消糜栓的提取工艺。方法 在单因素实验的基础上,以药材粉碎粒度、料液比、浸泡时间和提取时间为自变量,以浸膏得率和盐酸小檗碱、没食子酸含量为因变量,用Box-Behnken法设计不同水平实验,用Design-Expert 8.0.6.1软件建立二次回归模型,优化提取工艺。结果 最佳提取工艺:粉碎度为中粉、料液比为1∶19、浸泡时间为12 h、提取时间为2 h,此时妇科消糜栓中有效成分没食子酸含量为9.92%、浸膏得率为39.35%、盐酸小檗碱含量为10.98%、综合评分为25.98%。结论 响应面法优化所得提取工艺参数稳定可行,可在实际生产中应用。Objective To optimize the extraction process of traditional preparation,Fuke Xiaomi Suppository(FXS) by response surface methodology.Methods On the basis of single factor experiment,the particle size,solid-liquid ratio,soaking time and extraction time of medicinal materials were selected as independent variables,the yield of extract,berberine hydrochloride and gallic acid contents were taken as dependent variables.Box-behnken method was used to design the experiments at different levels.The quadratic regression model was established with Design-Expert 8.0.6.1 software to optimize the extraction process.Results The optimized extraction conditions were as follows:the crushing degree was medium powder,the ratio of solid to liquid was 1∶19,the soaking time was 12 hours,and the extraction time was 2 hours.Under these conditions,the content of gallic acid was 9.92%,the yield of extract was 39.35%,the content of berberine hydrochloride was 10.98%,and the comprehensive score was 25.98%.Conclusion The extraction parameters optimized by response surface method are stable and feasible,which can be used in practical production.
