摘  要:目的 探究紫花地丁总黄酮(total flavones from Viola yedoensis Makino,TFV)调控Toll受体3(toll-like receptor 3,TLR3)、β干扰素TRI结构域衔接蛋白(TIR-domain containing adaptor inducing interferon-β,TIRF)通路对单纯疱疹病毒性脑炎(herpes simplex virus en-cephalitis,HSE)的作用机制。方法 取50只SPF小鼠随机分为空白对照组、TFV组、Ⅰ型单纯疱疹病毒(herpes simplex virus-1,HSV1)-生理盐水(normal saline,NS)组、HSV-阿昔洛韦(acyclovir,ACV)组和HSV1-TFV组,每组10只。空白对照组、TFV组不做处理,其余3组注射HSV1病毒建立小鼠HSE模型(LD_(50),50×10^(-4)/mL^(-1))。造模后第1天,TFV组给予0.15 mL TFV灌胃(质量浓度50μg·mL^(-1)),HSV1+NS组给予NS 0.15 mL灌胃,HSV1+ACV组给予ACV 0.15 mL灌胃(质量浓度60μg·mL^(-1)),HSV1+TFV组给予TFV 0.15 mL灌胃(质量浓度50μg·mL^(-1))。HE染色观察脑组织的病理变化,ELISA法检测INF-α、IL-6的含量,Western blot检测TLR3、TRIF蛋白的表达水平,RT-PCR法检测TLR3、TRIF mRNA的表达水平。结果 与HSV1+NS组相比,HSV1+ACV组及HSV1+TFV组小鼠脑组织肿胀减轻,神经细胞变性减轻,轻微出血,炎性细胞明显减少,INF-α、IL-6含量及TLR3、TRIF mRNA和蛋白的表达水平明显降低(P<0.05),而空白对照组与TFV组各指标的差异无统计学意义。结论 TFV可减轻HSE小鼠炎性反应,改善脑组织损伤,可能与调控TLR3/TRIF通路有关。Objective To explore the effect of total flavones from Viola yedoensis Makino(TFV)on herpes simplex virus(HSV)encephalitis in mice and mechanism on toll-like receptor 3(TLR3)and TIR-domain containing adaptor inducing interferon-β(TIRF)pathway.Methods 50 SPF mice were randomly divided into blank control group,TFV group,herpes simplex virus-1(HSV1)+normal saline(NS)group,HSV1+acyclovir(ACV)group,and HSV1+TFV group,each group with 10 mice.The blank control group and TFV group were left untreated,the other groups were injected with HSV1 virus to establish a mouse HSE model(LD50,50×10^(-4)·mL^(-1)).On the first day after modeling,the TFV group were given 0.15 mL TFV(mass concentration 50μg·mL^(-1)),the HSV1+NS group were given 0.15 mL NS,and the HSV1+ACV group were given 0.15 mL ACV(mass concentration 60μg·mL^(-1)),and the HSV1+TFV group were given 0.15 mL TFV(mass concentration 50μg·mL^(-1)).HE staining was used to observe the pathological changes of brain tissue,ELISA method was used to detect the cytokine interferon-α(INF-α)and interleukin-6(IL-6)content,Western blot was used to detect the TLR3 and TRIF protein expression,and RT-PCR method was used to detect the TLR3 and TRIF mRNA expression in each group.Results Compared with the HSV1+NS group,the mouse brain tissue swelling,nerve cell degeneration,bleeding and inflammatory cells of HSV1+ACV group and HSV1+TFV group were significantly reduced.The INF-α,IL-6,TLR3 and TRIF mRNA and protein expression levels were significantly reduced(P<0.05),but the blank control group and TFV group had no statistically significant difference(P>0.05).Conclusion TFV can reduce the inflammatory response in HSE mice and alleviate the brain tissue damage,which may be related to the regulation of TLR3/TRIF pathway.
