摘 要:为提高豆科-根瘤菌共生固氮的效率,研究中华根瘤菌HH103菌型各效应因子突变体对大豆结瘤的影响,以其效应因子NopAA、NopT和NopC为研究对象,采用三亲杂交的方法获得突变体HH103ΩNopAA/T/C,PCR鉴定后,以绥农14为材料进行结瘤表型分析,发现突变体HH103ΩNopAA/T/C可以影响大豆结瘤数量以及结瘤重量,但对瘤大小没有明显影响;降低平均结瘤数和瘤鲜重干重,与突变体HH103ΩNopC有显著差异。上述结果说明效应因子NopAA、NopT和NopC相互调控,在大豆-根瘤菌共生体系建立过程中发挥重要作用。To improve the nodulation effect,typeⅢeffect factor NopAA,NopT,and NopC of Sinorhizobium fredii strain HH103 were studied and mutant HH103ΩNopAA/T/C(by triparental hybridization)was obteined.The effect of each effector mutant on soybean nodulation was compared by nodulation phenotype identification.Soybean cultivar Suinong 14 was used as the material for nodulation phenotype analysis.It was found that the mutant HH103ΩNopAA/T/C could affect the nodulation number and nodulation weight of soybean,but had no significant ef⁃fect on nodule size.In terms of the average number of nodules,the mutant HH103ΩNopAA/T/C decreased the num⁃ber of nodules.In terms of fresh weight and dry weight,there was significant difference between mutant HH103ΩNopAA/T/C and mutant HH103ΩNopC,and mutant HH103ΩNopAA/T/C inhibited nodulation weight.It is suggested that the effect factor NopAA,NopT,and NopC play important roles in establishment of soybean-rhizo⁃bium symbiosis system.
