摘  要:目的探究电针不同腧穴对急性结肠炎症模型大鼠细胞焦亡通路NOD样受体蛋白3(NLRP3)/半胱氨酸天冬氨酸酶-1(Caspase-1)/白细胞介素-1β(IL-1β)信号通路的影响及不同腧穴对急性结肠炎症是否具有特异性。方法将36只SD大鼠随机分为空白组、模型组、各电针干预组(分别为天枢组、上巨虚组、足三里组、大肠俞组),每组6只。除空白组外,其余各组均采用冰乙酸灌肠法造模。给予各电针干预组电针治疗3 d,每天1次,留针20 min。苏木精-伊红(HE)染色法观察各组大鼠结肠组织病理改变;免疫组织化学(IHC)法检测NLRP3在各组大鼠结肠组织的阳性表达;ELISA测定大鼠血清中IL-1β的含量;Wsetern blotting测定大鼠结肠中NLRP3、Caspase-1的蛋白表达。结果与空白组相比,模型组大鼠血清中IL-1β含量显著升高(P<0.05);与模型组相比,各电针干预组大鼠血清中IL-1β含量均下降(P<0.05);足三里组与天枢组、大肠俞组、上巨虚组相比,IL-1β明显降低(P<0.05)。与空白组相比,模型组大鼠结肠组织中NLRP3蛋白表达显著增高(P<0.05);与模型组相比,足三里组NLRP3蛋白表达下降(P<0.05);与空白组相比,模型组大鼠结肠组织中Caspase-1蛋白表达显著升高(P<0.05),与模型组相比,各电针干预组中Caspase-1蛋白表达均降低(P<0.05)。结论电针治疗急性结肠炎症模型大鼠,可能通过抑制NLRP3/Caspase-1/IL-1β信号通路来改善大鼠结肠黏膜损伤,达到促进炎症修复的目的;电针天枢、上巨虚、足三里、大肠俞对急性结肠炎症大鼠均有所改善,电针足三里在降低NLRP3蛋白表达、抑制Caspase-1的表达及降低IL-1β的含量方面较其他电针干预组可能更为显著。Objective:To explore the effects of different acupoints on NOD-like receptor protein 3(NLRP3)/Caspase-1/interleukin-113(IL-1 p)signaling pathway of pyroptosis in acute colitis model rats,and whether different acupoints have specificity for acute colitis.Methods:36 SD rats were randomly divided into the blank group,model group and each electroacupuncture(EA)intervention group[EA intervention groups included Tianshu(ST 25)group,Shangjuxu(ST 37)group,Zusanli(ST 36)group and Dachangshu(BL 23)group],6 rats in each group.Except for the blank group,the rats in the other groups were induced by glacial acetic acid enema.EA was given to the EA intervention groups for 3 days,once a day,and the needles were retained for 20 min.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of colon tissue;immunohistochemistry(IHC)was used to detect the positive expression of NLRP3 in the colon tissue of rats in each group;ELISA was used to measure the content o£IL-lp in rat serum;Western blotting was used to detect the protein expression of NLRP3 and Caspase-1 in rat colon.Results:Compared with the blank group,the serum level of IL-lp in the model group was significantly increased(P<0.05);compared with the model group,the contents of IL-ip in the serum of the rats in each EA intervention group were decreased(P<0.05);compared with the Tianshu(ST 25)group,Dachangshu(BL 23)group,and the Shangjuxu(ST 37)group,the IL-lp level in the Zusanli(ST 36)group was significantly decreased(P<0.05);compared with the blank group,the expression of NLRP3 protein in the colon tissue of the model group was significantly increased(P<0.05);compared with the model group,the expression of NLRP3 protein in the Zusanli group was decreased(P<0.05);compared with the blank group,the expression of Caspase-1 protein in the colon tissue of the model group was significantly increased(P<0.05);compared with the model group,the expression of Caspase-1 protein was decreased in each EA intervention group(P<0.05).Conclusion:EA treatment for acute colit
