摘 要:为了研究牛血清白蛋白(BSA)对犬精液冷冻保存效果的影响,试验配制含有不同浓度(0、3%、5%、7%)BSA的精液冷冻稀释液用于保存犬精液,测定冷冻-解冻后精子活力、质膜完整率、顶体完整率、总抗氧化能力(T-AOC)、丙二醛(MDA)含量及线粒体膜电位(MMP)等指标。结果表明:3%和5%BSA组冷冻-解冻后精子活力、质膜完整率、顶体完整率和T-AOC均显著高于对照组(P<0.05);3%BSA组MDA含量显著低于其他各组(P<0.05),线粒体膜电位值显著高于其他各组(P<0.05);除精子活力外,7%BSA组和对照组之间各项指标均差异不显著(P>0.05)。说明在犬精液冷冻稀释液中添加BSA能有效改善冷冻-解冻后精液品质,对提高精子抗氧化能力及代谢能力都有显著效果,BSA的最适添加浓度为3%。To study the effects of bovine serum albumin(BSA)on the cryopreservation of canine semen,cryopreservation diluents containing BSA with levels of 0,3%,5%and 7%,respectively,were prepared for freezing canine semen.The sperm motility,plasma membrane integrity,acrosome integrity,total antioxidant capacity(T-AOC),malondialdehyde(MDA)content and mitochondrial membrane potential(MMP)were assessed after freezing and thawing.The results showed that the sperm motility,plasma membrane integrity,acrosome integrity and T-AOC in 3%and 5%BSA groups were improved than those in control group(P<0.05).The MDA content in 3%BSA group was lower than that of other groups(P<0.05),the MMP in 3%BSA group was significantly higher than other groups(P<0.05).Except for sperm motility,there was no significant difference between the 7%BSA group and the control group(P>0.05).In conclusion,the addition of BSA to the semen dilution could effectively improve the quality of frozen-thawed canine semen,and might have a positive effect on improving the antioxidant capacity and metabolic capacity of semen,and the optimal addition concentration of BSA would be 3%.
