武当山地区山慈菇药材质量研究

摘　　要：目的:建立武当山地区山慈菇的质量标准,以便为其质量控制提供参考。方法:采用性状和显微鉴别的方法对药材的特征进行观察描述,采用薄层色谱法对药材进行定性鉴别,参照《中国药典》相关方法测定其灰分和浸出物的含量,并使用紫外分光光度法测定山慈菇不同生长年份总黄酮、总酚、总生物碱的含量。结果:山慈菇大多呈圆球或圆锥形,显微鉴别可依据草酸钙针晶长短、导管种类及大小、淀粉粒大小等特征;两批次山慈菇的总灰分含量分别为4.78%、4.53%,酸不溶性灰分含量为0.87%、1.28%;水溶性浸出物(冷浸、热浸法)的平均含量分别为10.9%、7.3%,醇溶性浸出(冷浸、热浸法)的平均含量分别为2.0%、3.8%;一年生的山慈菇总黄酮、总酚、总生物碱含量最高,分别为4.695、7.408、0.605 mg/g。结论:上述鉴别和含量测定的方法操作简便、准确,可用于武当山慈菇的质量控制。Objective To establish the quality standard of Cremastra appendiculata(D.Don)Makino in Wudang mountain area to provide the basis for its quality control.Methods The characteristics of the medicinal materials were observed and described by the method of character and microscopic identification,and the qualitative identification of the medicinal mate⁃rials was carried out by thin-layer chromatography.The content of ash and extracts were determined by referring to the rele⁃vant methods of"Chinese Pharmacopoeia",and the contents of total flavonoids,total phenols and total alkaloids in differ⁃ent growth years were determined by UV spectrophotometry.Results Most C.appendiculatas are mostly spherical or conical.Microscopic identification can be made from the length of calcium oxalate needle,the type and size of the catheter,the size of starch grain and other characteristics.The total ash content of the two batches was 4.53%and 4.78%,the acid-insoluble ash content was 0.87%and 1.28%,the average content of water-soluble extract(cold and hot leaching)was 10.9%and 7􀆰3%,and the average content of alcohol-soluble leaching(cold and hot leaching)was 2.0%and 3.8%.The contents of total flavonoids,total phenols and total alkaloids of C.appendiculata in one year were the highest,which were 4.695,7􀆰408,0.605 mg/g,respectively.Conclusion The above methods for identification and content determination are simple and accurate,and can be used for quality control of the C.appendiculata in Wudang mountain area.
