摘 要:目的:建立柴胡药材的HPLC检测方法以评价不同地区柴胡药材的质量。方法:采用HPLC法测定10批不同产地柴胡中柴胡皂苷a、d的含量。色谱条件如下:色谱柱为日本岛津inertsilODS-3色谱柱(5μm,4.6 mm×250 mm),流动相为乙腈-水(45∶55)等度洗脱,流速1.0 min·mL^(-1),检测波长203 nm,柱温30℃。结果:柴胡皂苷a的标准曲线回归方程:Ya=412288X+1026.4(r2=0.9998),柴胡皂苷d的标准曲线回归方程:Yd=443285X-10770(r^(2)=0.9998),建立的HPLC测定方法简单易行,精准可靠。所测10批柴胡药材含量均符合药典要求,综合评分分析显示四川和西藏的柴胡饮片得分较高。结论:10批柴胡药材整体质量良好,四川、西藏的自然环境和气候类型可能更适宜柴胡生长,柴胡皂苷含量更高。Objective To establish an HPLC detection method for bupleuri radix to evaluate the quality of bupleurum radix from different regions.Methods HPLC method was used to determine the content of saikosaponins a and d in 10 batches of Bupleurum from different origins.The chromatographic conditions were as follows:the chromatographic column was Japan Shimadzu inertsil ODS-3 chromatographic column(5μm,4.6 mm×250 mm),the mobile phase was acetonitrile water(45∶55)with equal elution,and the flow rate was 1.0 min·mL^(-1),detection wavelength 203 nm,column temperature 30℃.Results Standard curve regression equation for saikosaponin a was Ya=412288X+1026.4(r^(2)=0.9998),and standard curve regression equation for saikosaponin d was Yd=443285X-10770(r^(2)=0.9998).The established HPLC determination method is simple,feasible,accurate,and reliable.The content of 10 batches of Bupleurum medicinal herbs tested met the requirements of the pharmacopoeia,and comprehensive score analysis showed that the scores of Bupleurum decoction pieces in Sichuan and Tibet were relatively high.Conclusion The overall quality of the 10 batches of Bupleurum medicinal herbs is good,and the natural environment and climate types in Sichuan and Tibet may be more suitable for the growth of Bupleu⁃rum,with a higher content of saikosaponins.
