摘 要:目的:观察睡菜水提液对大鼠的急性毒性.方法:预实验:灌胃给予小鼠水提液剂量分别为2455、1841.25、1380.94、1035.70 g/L,观察给药当天及7 d内小鼠的状态,计算半数致死量(50%lethal dose,LD_(50));最大耐受量实验:灌胃给予睡菜最大剂量2455 g/L,观察给药后14 d内小鼠状态和死亡情况,给药14 d后检测小鼠的血液生化指标、计算主要脏器指数以及观察主要脏器病理学变化.结果:未能测出睡菜水提液的半数致死量.最大耐受量实验显示:给药后14 d内小鼠状态良好,无中毒反应或死亡情况.给药组小鼠第3、7、11、14 d体质量增长与空白组比较无统计学差异(P>0.05);给药组小鼠血液生化指标、主要脏器指数与空白组无统计学差异(P>0.05);组织形态学、组织病理学检查未发现睡菜水提液对小鼠主要脏器产生病理改变.结论:睡菜水提液的最大无毒剂量大于2455 g/L,是成人临床用药(15 g)的100.6倍,是安全的用药剂量范围.Objective:To observe the acute toxicity of Menyanthes trifoliata L in mice.Methods:Pre-test:mice were given the aqueous extract at doses of 2455,1841.25,1380.94 and 1035.70 g/L by gavage,and the status of mice was observed on the day of administration and within 7 d.The LD_(50)(50% lethal dose) was calculated;the maximum tolerance test:The mice were given 2455 g/L by gavage,and the status and death of mice were observed after the administration of the drug within 14 d.Results:The LD_(50) values of the aqueous extract of sleepy vegetable could not be measured.The maximum tolerance test indicated that the mice were in good status within 14 d after administration,and there was no toxic reaction or death.There was no statistical difference in the increase of body mass of mice in the 3rd,7th,11th and 14th day of administration compared with the control group(P>0.05);there were no statistical difference in the blood biochemical index and main organ index of mice in the administration group compared with the control group(P>0.05);the histomorphology and histopathology examination did not reveal any pathological changes in the main organs of mice caused by the aqueous extract of water hyacinth.Conclusion:The maximum non-toxic dose of aqueous extract of water hyacinth is greater than 2455 g/L,which was 100.6(15 g) times of the clinical dose for adults and was a safe dose range.
