摘 要:探究超声协同酶解法优化光果莸中黄酮的提取工艺,测定野黄芩苷含量,研究光果莸不同部位体外抗血小板聚集活性。以黄酮提取率为考察指标,通过单因素实验与正交实验设计分析各因素显著性,正交实验得出光果莸黄酮最佳提取条件;采用HPLC测定光果莸不同部位的野黄芩苷含量,以血小板聚集曲线为指标,考察光果莸不同部位活性成分的体外抗血小板聚集作用。光果莸黄酮提取最佳优化条件为酶解时间100 min,酶解温度65℃,提取加酶量60 mg,V(提取液)∶m(光果莸粉末)=10∶1 mL/g,超声功率600 W,超声时间50 min;花、叶、茎、根中w(野黄芩苷)分别为5.42、9.67、0.97、1.07μg/g;在抗血小板聚集作用研究中,光果莸叶的抑制率最高,达到65.96%。超声协同酶解法优化光果莸黄酮提取工艺简单、可行,体外抗血小板聚集活性较高,与其功效吻合,为后期有效开发光果莸提供了一定的理论基础。The extraction process of flavonoids from Caryopteris tangutica by ultrasonic assisted enzymatic hydrolysis was optimized,and the content of baicalin was determined.The antiplatelet aggregation activity of different parts of Caryopteris tangutica in vitro was analyzed.Taking the extraction rate of flavonoids as the index,the significance of each factor was analyzed by single factor test and orthogonal test design,and the optimal extraction conditions of flavonoids from Caryopteris tangutica were obtained by orthogonal design.The content of scutellarin in different parts of Caryopteris tangutica was determined by HPLC.The platelet aggregation curve was used as an index to investigate the in vitro antiplatelet aggregation effect of active components from different parts of Caryopteris tangutica.The optimal extraction conditions of flavonoids from Caryopteris tangutica were as follows:the enzymatic hydrolysis time was 100 min,the enzymatic hydrolysis temperature was 65℃,the amount of enzyme added for extraction was 60 mg,the ultrasonic liquid material ratio was 10∶1,the ultrasonic power was 600 W,and the ultrasonic time was 50 min.The content of scutellarin was 5.42μg/g for flowers,9.67μg/g for leaves,0.97μg/g for stems and 1.07μg/g for roots.In the study of antiplatelet aggregation,the inhibitory rate of the leaves of Caryopteris tangutica was the highest,reaching 65.96%.The optimization of the extraction process of flavonoids from Caryopteris tangutica by ultrasonic synergistic enzymatic hydrolysis is simple and feasible.The anti platelet aggregation activity in vitro is high,which is consistent with its efficacy.It provides a certain theoretical basis for the effective development of Caryopteris tangutica.
