摘 要:测定了中国4个细胞源猪瘟活疫苗(C株)E2基因全序列,将其与GenBank上13个猪瘟病毒的E2基因序列(含C株原始毒株、7个猪瘟病毒C株以及国际上5个主要猪瘟活疫苗毒株的E2基因全序列)进行了核苷酸序列、推导的氨基酸序列进行遗传变异分析,通过生物信息学手段对预测的E2蛋白N-糖基化位点、E2蛋白磷酸化位点和理化特性以及模拟的蛋白空间结构进行了比较,并通过免疫攻毒试验对4个细胞源猪瘟疫苗(C株)的免疫效果进行了验证。结果表明,与猪瘟病毒C株原始种毒相比,尽管中国4个细胞源猪瘟活疫苗的E2基因核苷酸序列、推导的E2蛋白氨基酸序列或模拟的三维结构等发生个别核苷酸变异、个别氨基酸位点突变或缺失,但均不影响猪瘟C株的免疫原性,猪瘟疫苗C株仍然是防控猪瘟的最有效武器。The complete sequences of E2 gene of four cell derived live classical swine fever vaccines(strain C)in China were determined.The nucleotide sequence and deduced amino acid sequence ofE2 gene of 13 classical swine fever viruses(including the complete sequences of E2 gene of original strain C,7 classical swine fever virus strains C and 5 major classical swine fever vaccine strains in the world)on GenBank were analyzed for genetic variation.The predicted N-glycosylation sites The phosphorylation sites,physical and chemical properties of E2 protein and the simulated protein space structure were compared,and the immune effects of four cell derived classical swine fever vaccines(strain C)were verified by immune challenge test.The results showed that,compared with the original strain of classical swine fever virus C,although the nucleotide sequence of E2 gene,deduced amino acid sequence of E2 protein or simulated three-dimensional structure of the four cell derived live classical swine fever vaccines in China had individual nucleotide variation,mutation or deletion of individual amino acid sites,they did not affect the immunogenicity of classical swine fever virus C strain,and classical swine fever vaccine C strain was still the most effective weapon for the prevention and control of classical swine fever.
