摘  要:[目的]通过探究茶多酚、白藜芦醇、丁香酚、氟康唑4种化合物对杧果炭疽病菌(Colletotrichum gloeosporioides)的抑菌活性及对病原菌漆酶基因家族成员转录水平的影响,为该家族在寻找杧果炭疽病菌潜在抑菌化合物提供参考。[方法]通过菌丝生长速率法测定4种抑菌化合物对杧果炭疽病菌菌丝生长、分生孢子萌发和附着胞形成、黑色素合成的影响;同时测定这4种抑菌化合物对13个漆酶家族成员基因表达量和对胞外漆酶酶活的影响。[结果]在菌丝生长方面,茶多酚、白藜芦醇、丁香酚和氟康唑对杧果炭疽病菌菌丝生长的EC_(50)分别为15010、1109.88、308.26和163.52(mg·L^(-1))。从分生孢子萌发率和附着胞形成率来看,这4种抑菌化合物均对分生孢子萌发起促进作用,而都抑制了附着胞的形成。在黑色素合成影响来看,茶多酚和白藜芦醇均抑制黑色素合成,而丁香酚和氟康唑均促进黑色素合成。qRT‐PCR结果显示,100 mg·L^(-1)的茶多酚和168 mg·L^(-1)的白藜芦醇对2/13漆酶家族成员转录调控有抑制作用,164 mg·L^(-1)的丁香酚和306 mg·L^(-1)的氟康唑都对3/13漆酶家族成员转录调控有抑制作用。100 mg·L^(-1)的茶多酚和168 mg·L^(-1)的白藜芦醇、164 mg·L^(-1)丁香酚和306 mg·L^(-1)氟康唑均对杧果炭疽病菌漆酶酶活有显著抑制作用。[结论]这4种抑菌化合物都对杧果炭疽病菌的菌丝生长、附着胞形成、胞外漆酶酶活有显著抑制作用,对分生孢子萌发有促进作用,而对黑色素合成、家族成员转录调控有所差异。[Objective]By investigating the antibacterial activities of four compounds,namely tea polyphenols,resveratrol,eugenol,and fluconazole,on Colletotrichum gloeosporioides and their effects on the transcrip⁃tional levels of the laccase gene family members of the pathogenic bacteria,this study aimed to provide a refer⁃ence for finding potential antibacterial compounds in this family for the control of C.gloeosporioides.[Meth⁃ods]The mycelial growth rate method was used to determine the effects of four bacteriostatic compounds on the hyphae growth,conidia germination,appressorium formation,and melanin synthesis of C.gloeosporioi⁃des.The effects of four bacteriostatic compounds on the expression levels of 13 laccase gene family members and on the extracellular laccase enzyme activity were also measured.[Results]The results showed that EC_(50) values of tea polyphenols,resveratrol,eugenol,and fluconazole were 15010,1109.88,308.26,and 163.52 mg·L^(-1),respectively,on the inhibition of the growth rate of mycelium.In terms of spore germination and appressorium formation,all four antibacterial compounds promoted spore germination but inhibited ap⁃pressorium formation.Regarding melanin synthesis,tea polyphenols and resveratrol inhibited the synthesis rate while eugenol and fluconazole promoted melanin synthesis.qRT⁃PCR results showed that 100 mg·L^(-1) of tea polyphenols and 168 mg·L^(-1) of resveratrol had inhibitory effects on the transcriptional regulation of 15%laccase gene family members,and 164 mmg·L^(-1) of eugenol and 306 mg·L^(-1) of fluconazole had inhibitory ef⁃fects on the transcriptional regulation of 23%laccase gene family members.Tea polyphenols at 100 mg·L^(-1),resveratrol at 168 mg·L^(-1) eugenol at 164 mg·L^(-1),and fluconazole at 306 mg·L^(-1) all significantly inhibited the extracellular laccase enzyme activity of C.gloeosporioides.[Conclusion]All four antibacterial compounds have significant inhibitory effects on the hyphae growth,appressorium formation,and extracellular laccase en�
