摘  要:以甘肃民乐紫皮大蒜为试验材料,通过茎尖离体培养和激素配比试验,筛选最佳培养基,获得无病毒组培苗。结果表明,切取0.3~0.5mm+叶原基1片的茎尖外植体为标准,筛选出MS为茎尖基础启动培养基,成活率和成苗率均达90.0%以上;以MS+0.5mg/L~1.0mg/L6-BA+0.05mg/L~0.10mg/L NAA为增殖培养基,增殖倍数在3.11~4.25;以MS+NAA0.6mg/L为最适宜生根培养基,生根率最高达92%,茎尖脱毒率达94%,形成了甘肃民乐紫皮大蒜脱毒快繁关键技术体系。为提纯复壮地方特色品种提供了重要技术途径。Using Gansu Minle purple-skinned garlic as the test material,the best medium was screened by stem tip ex vi⁃vo culture and hormone ratio test to obtain virus-free histopathic seedlings.The results showed that MS was selected as the ba⁃sic starter medium for stem tips,and the survival rate and seedling establishment rate were above 90.0%;MS+0.5mg/L~1.0mg/L6-BA+0.05mg/L~0.10mg/L NAA was used as the proliferation medium,and the proliferation multiplier was between 3.11~4.25.The most suitable rooting medium was MS+NAA0.6mg/L.The rooting rate reached 92%,and the rate of stem tip detoxifica⁃tion reached 94%,forming a key technology system for detoxification and fast propagation of Gansu Minle purple garlic.It has provided an important technical way to purify and strengthen the local characteristic varieties.
