摘  要:目的:建立河西走廊气候影响下的过敏性哮喘大鼠模型(即河西走廊气候特点的过敏性哮喘大鼠模型),探讨河西走廊气候因素对过敏性哮喘大鼠模型气道炎症的影响。方法:用卵蛋白致敏SD大鼠的方法建立过敏性哮喘模型,将过敏性哮喘模型暴露于河西走廊气候环境下,将SD大鼠按体重均衡原则随机分为空白组、哮喘模型组、河西走廊气候环境暴露低、中、高剂量组,光镜下观察肺组织的病理学变化,用ELISA法检测大鼠血浆中TNF-α和IL-4的水平。结果:肺组织病理学显示,哮喘模型组肺泡和支气管结构不完整,肺泡间隔明显增厚,肺泡壁毛细血管扩张充血,肺泡腔内和支气管壁内可见大量炎性细胞浸润,支气管上皮中杯状细胞增多。随着河西走廊气候因素暴露程度的加重,肺组织损伤程度加重,肺泡逐渐出现实变和出血,支气管内有分泌物潴留和出血。大鼠血浆TNF-α和IL-4水平显示,与空白组相比,各组大鼠血浆TNF-α水平均降低、IL-4水平均升高、TNF-α/IL-4比值均降低,差异均具有统计学意义(P<0.05或P<0.01);与模型组相比,河西走廊气候因素暴露各组大鼠血浆TNF-α水平均降低,IL-4水平均升高,TNF-α/IL-4比值均降低,差异均具有统计学意义(P<0.01)。河西走廊气候因素暴露各组两两相比,血浆TNF-α水平依次降低、IL-4水平依次升高、TNF-α/IL-4比值依次降低,差异均具有统计学意义(P<0.01)。结论:随着河西走廊气候因素暴露程度的加重,哮喘大鼠模型气道损伤和炎症程度加重,其可能的作用机制是上调IL-4水平和下调TNF-α水平有关。Objective:To establish an allergic asthma rat model under the influence of the climate of the Hexi Corridor(i.e.an allergic asthma rat model characterized by the climate of the Hexi Corridor)and to investigate the effects of climatic factors of the Hexi Corridor on the airway inflammation of the allergic asthma rat model.Methods:The allergic asthma model was established by the method of ovalbumin sensitization of SD rats,and the allergic asthma model was exposed to the climatic environment of the Hexi Corridor.SD rats were randomly divided into blank group,asthma model group,and low,medium and high dose groups ex⁃posed to the climatic environment of the Hexi Corridor according to the principle of weight balance,and the pathological changes of lung tissues were observed under light microscope,and the plasma levels of TNF-αand IL-4 levels by ELISA.Results:Lung histopathology showed that the alveolar and bronchial structures of the asthma model group were incomplete,the alveolar septa were significantly thickened,the alveolar wall capillaries were dilated and congested,a large number of inflammatory cells were seen in⁃filtrating the alveolar lumen and the bronchial wall,and the cupular cells in the bronchial epithelium were increased.As the degree of exposure to climatic factors in the Hexi Corridor increased,the degree of lung tissue damage worsened and the alveoli gradually became realistic and haemorrhagic,with secretion retention and haemorrhage in the bronchi.Plasma TNF-αand IL-4 levels in rats showed that compared with the blank group,plasma TNF-αlevels decreased,IL-4 levels increased and TNF-α/IL-4 ratio de⁃creased in all groups,and the differences were statistically significant(P<0.05 or P<0.01);compared with the model group,plas⁃ma TNF-αlevels decreased and IL-4 levels increased in all groups exposed to climate factors in the Hexi Corridor.Compared with the model group,plasma TNF-αlevels decreased,IL-4 levels increased and TNF-α/IL-4 ratio decreased in all groups,and the differences were stat
