摘  要:研究旨在运用高效液相色谱法(HPLC)建立饲用植物蒲公英水提物的特征图谱,并用聚类分析和主成分分析对蒲公英水提物进行质量评价。采用Sunfire C18色谱柱,以乙腈-0.1%磷酸水为流动相,流速为0.8 m L/min,进样量10μL,柱温35℃,检测波长329 nm,将22批蒲公英水提物色图谱导入中药色谱指纹图谱相似度评价系统软件(2012年版)中,选取分离度、峰型较好的8个峰为特征峰,得到蒲公英水提物特征图谱,并进行相似度评价、聚类分析和主成分分析。结果表明:试验建立的蒲公英水提物HPLC特征图谱共确定了8个特征峰,指认了单咖啡酰酒石酸、绿原酸、咖啡酸和菊苣酸4个色谱峰,相似度结果为0.869~0.999,说明22批蒲公英水提物质量相对稳定;当欧氏距离为4时,聚类分析将22批不同产地的蒲公英分成3类,S6一类,S4一类,其余批次为一类;主成分分析中单咖啡酰酒石酸和菊苣酸具有最大的贡献度。通过相似度评价、聚类分析和主成分分析与特征图谱相结合,可为蒲公英水提物质量评价提供参考。The aim of this study was to establish the characteristic chromatogram of Taraxacum mongoli⁃cum aqueous extract(TMAE)by high performance liquid chromatography(HPLC)and evaluate its quali⁃ty by cluster analysis(CA)and principal component analysis(PCA).The column was Sunfire C18 with acetonitrile-0.1%phosphoric acid water as the mobile phase,the flow rate was 0.8 mL/min,the injection volume was 10μL,the column temperature was 35℃,and the detection wavelength was 329 nm.The chromatograms of 22 batches of TMEA were imported into the similarity evaluation system software for Chinese medicine chromatography(2012 version),and the 8 peaks with good separation and peak shape were selected as the characteristic peaks to obtain the characteristic chromatograms of TMEA.Simi⁃larity evaluation,CA and PCA were carried out using the peak areas of the shared characteristic peaks normalized as variables.The results showed that:a total of 8 shared peaks were identified in the HPLC characteristic chromatograms of the aqueous extract of dandelion established in the tri⁃al,and four peaks were identified for caftaric acid,chlorogenic acid,caffeic acid and chicory acid.The similarity results were 0.869-0.999,indicating that the 22 batches of TMAE samples were stable.When the Euclidean distance was 4,the CA classified 22 batches of Taraxacum mongolicum of different ori⁃gins into three categories,S6 into one category,S4 into one category and the rest of the batches into one category.Caftaric acid and chlorogenic acid had the greatest contribution in PCA.The combination of similarity evaluation,CA and PCA with characteristic chromatogram can provide a reference for the quali⁃ty evaluation of TMEA.
