摘  要:目的:基于LC-MS寻找儿童ITP潜在生物标记物。方法:采集患儿及健康儿童外周血进行LC-MS检测,通过统计学方法分析,经数据库鉴定代谢物结构。结果:LC-MS/MS检测获得原始数据,经主成分分析、层次聚类分析、秩和检验及HMDB数据库中匹配鉴定,再通过ROC曲线分析,最终筛选鉴定出5种潜在生物标志物:间甲酚、腐胺、5-羟色胺、2-氧代精氨酸、精氨基琥珀酸。结论:检测鉴定出的5种关键差异性代谢物可能可以作为鉴别ITP患儿和健康儿童的临床实验室生物标记物。Objective:To find the potential biomarkers of ITP based on LC-MS.Methods:The peripheral blood of the patient and healthy children was collected for LC-MS detection,and the metabolite structure was identified by database.Results:The original data obtained by LC-MS/MS were identified by principal component analysis,hierarchical cluster analysis,rank sum test and HMDB database matching,and then by ROC curve analysis,5 potential biomarkers were finally screened and identified:m-cresol,putreamine,5-hydroxytryptamine,2-oxo-arginine,argininosuccinic acid.Conclusionl:The five key differential metabolites identified may be used as clinical laboratory biomarkers to distinguish ITP children from healthy children.
