摘  要:为了观察五味子枸杞子大枣复合醋(简称五味子复合醋,SCV)对大鼠非酒精性脂肪肝(NAFLD)的保护作用并探讨其作用机制,本试验将50只雄性SD大鼠随机分为2个组,分别为对照组(10只,正常饮食)和NAFLD模型组(40只,高脂饮食),于第4周末将NAFLD模型组大鼠随机分为4个组,分别为模型组(高脂饮食,给予蒸馏水)和SCV低、中、高剂量组(高脂饮食,分别给予25%SCV、50%SCV、100%SCV),于第8周结束试验;继而比较各组大鼠体重指数、血脂水平、肝脏功能、肝脏抗氧化能力,并通过蛋白质印迹法检测大鼠肝脏中脂质代谢PPARα/ACOX1/CPT1A通路和抗氧化Keap-1/Nrf2/HO-1通路相关蛋白的表达水平。结果显示,与模型组相比,SCV显著降低大鼠体重指数(P<0.01)、外周血和肝脏中甘油三酯(TG)和胆固醇(TC)含量(P<0.05或P<0.01)、外周血中低密度脂蛋白(LDL-C)含量、动脉粥样硬化指数(AI)(P<0.05或P<0.01)和血清转氨酶活性(P<0.05或P<0.01),极显著升高外周血中高密度脂蛋白(HDL-C)含量(P<0.01);且SCV可提高大鼠肝脏超氧化物歧化酶(SOD)活性(P<0.05)并降低丙二醛(MDA)含量(P<0.05),提高抗氧化能力;SCV还可减轻肝脏组织病理损伤;此外,SCV能显著提升大鼠肝脏PPARα、ACOX1、CPT1A、Nrf2和HO-1蛋白的表达水平(P<0.05或P<0.01),并显著降低Keap-1蛋白表达水平(P<0.05或P<0.01)。结果表明,SCV对大鼠NAFLD具有显著的保护作用,其分子机制可能与SCV调节PPARα/ACOX1/CPT1A和Keap-1/Nrf2/HO-1两条通路的相关蛋白表达水平相关。In order to assess the protective effect of Schisandra compound vinegar(SCV),consisting of Schisandra,Fructus lycii,Chinese date and vinager,on nonalcoholic fatty liver disease(NAFLD)in rats and explore its mechanism,this study randomly divided 50 male SD rats into two groups,the control group(10 rats,normal diet)and the NAFLD model group(40 rats,high-fat diet).At the end of the 4th week,NAFLD model rats were further divided into four groups,model group(high-fat diet,distilled water),SCV low-dose group(high-fat diet,25%SCV),SCV medium-dose group(high-fat diet,50%SCV)and SCV high-dose group(high-fat diet,100%SCV),with the test ending in the 8th week.The body mass index,blood lipid level,liver function and antioxidant capacity of rats in each group were compared,and the expression level of proteins involved in lipid metabolism(PPARα/ACOX1/CPT1A pathway)and antioxidant defense system(Keap-1/Nrf2/HO-1 pathway)in rat liver was detected by Western blot.The results showed that compared with model group,SCV significantly reduced the body mass index(P<0.01),triglyceride(TG)and total cholesterol(TC)content in peripheral blood and liver(P<0.05 or P<0.01),low-density lipoprotein cholesterol(LDL-C)content in peripheral blood,atherosclerosis index(AI)(P<0.05 or P<0.01)and serum transaminase activity(P<0.05 or P<0.01)in rats,significantly increased the high-density lipoprotein cholesterol(HDL-C)content in peripheral blood(P<0.01),significantly improved the antioxidant capacity of the body,due to its ability to increase superoxide dismutase(SOD)activity(P<0.05)and reduced malondialdehyde(MDA)content(P<0.05)in rat liver.SCV also reduced the pathological damage of liver tissue.In addition,SCV significantly improved the expression of PPARα,ACOX1,CPT1A,Nrf2 and HO-1 protein(P<0.05 or P<0.01)and reduced the expression of Keap-1(P<0.05 or P<0.01)in rat liver.Thus,SCV has a significant protective effect on NAFLD model rats,and the molecular mechanism may be related to the regulation of protein expression levels of both PPARα/ACOX1/C
