摘 要:为了探究槲皮素对慢性镉中毒小鼠肝、肾是否具有保护作用,本试验选用40只2月龄的雄性健康昆明小鼠随机分为4个组,对照组隔天腹腔注射生理盐水,槲皮素组隔天灌胃槲皮素[50 mg/(kg·bw)],镉中毒组隔天腹腔注射氯化镉[1 mg/(kg·bw)],槲皮素干预镉中毒组隔天灌胃槲皮素并腹腔注射氯化镉,持续1个月后,将小鼠处死,采集小鼠血液,生物化学法检测肝肾功能生化指标丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和尿素氮(BUN)水平,并采集肝、肾组织进行宏观和微观病理学观察。结果显示:与对照组相比,镉中毒组小鼠ALT、AST活力和BUN含量均升高,但差异均不显著(P>0.05);与镉中毒组相比,槲皮素组和槲皮素干预镉中毒组小鼠ALT、AST活力和BUN含量均降低,但差异均不显著(P>0.05)。与对照相比,槲皮素组肝、肾组织的颜色和形态结构无明显变化,镉中毒组肝、肾组织外观肿大,颜色暗红,质地易碎,病理切片可见细胞排列杂乱且不规则,部分细胞核溶解,细胞质空泡化;与镉中毒组相比,槲皮素干预镉中毒组肝、肾组织外观无明显异常变化,病理切片可见肝、肾细胞结构、边界和排列逐渐清晰规则,细胞核、细胞质的病变都明显改善。结果表明,槲皮素对慢性镉中毒小鼠导致的肝肾损伤具有一定的保护作用。In order to investigate the protective effect of quercetin on cadmium-induced hepato-nephrotoxicity of mice,this study divided a total of 40 male Kunming mice of clean grade into four groups:control group(intraperitoneal injection with saline every other day),quercetin group[50 mg/(kg·bw)quercetin gavage every other day],cadmium group[intraperitoneal injection with 1 mg/(kg·bw)cadmium chloride every other day],quercetin intervening cadmium group(quercetin gavage and intraperitoneal injection with cadmium chloride every other day).After one month,all mice were sacrificed and blood samples were collected.Indexes of liver and kidney function including ALT,AST and BUN levels were determined by biological chemistry method,and liver and kidney tissues were examined by macroscopic and microscopic histopathology.The results showed that compared with the control group,the levels of ALT,AST and BUN in cadmium group were increased,but the difference was not significant(P>0.05);compared with the cadmium group,the levels of ALT,AST and BUN were decreased in the quercetin group and the quercetin intervening cadmium group,but the difference was not significant(P>0.05).Compared with the control group,the color and morphology of liver and kidney in the quercetin group had no significant changes;the liver and kidney tissues in the cadmium group manifested swelling,dark red color,fragile texture;pathological sections revealed disordered and irregular cell arrangement,dissolved nucleus,and vacuolization in cytoplasm;compared with the cadmium group,no other obvious changes were observed in the quercetin intervening cadmium group,pathological sections revealed that the clear and regular structure of liver and kidney cells was present,and the lesions of the nucleus and cytoplasm were significantly improved.Thus,quercetin appears to improve and protect hepato-nephrotoxicity in mice caused by chronic cadmium poisoning.
