摘  要:目的:基于网络药理学的研究方法,探讨络石藤治疗类风湿关节炎(RA)的可能作用机制。方法:以口服生物利用度(OB)≥30%、类药性(DL)≥0.18为指标,通过中药系统药理学数据库与分析平台(TCMSP)数据库获取络石藤的活性成分、作用靶点,利用uniprot数据库将相应靶点蛋白转换为标准蛋白基因名。在Disgent数据库中检索“Rheumatoid arthritis”,获取RA疾病靶点运。利用韦恩图,得到药物成分和RA共同靶点,上传至Cytoscape 3.9.1构建“络石藤-化合物-靶点-类风湿关节炎”的网络图,并进行蛋白互作网络分析(PPI网络分析),GO富集和KEGG通路富集分析。结果:获取络石藤9种成分及109个作用靶点,RA疾病2722个相关靶点,络石藤-RA共同靶点57个,GO功能富集分析显示394个生物过程(BP)、41个细胞组成(CC)、70个分子功能(MF)。KEGG通路富集分析显示132条通路。结果:该研究初步揭示了青风藤中以青藤碱为主的6种有效活性成分发挥了抗RA的作用,治疗的关键靶点与AKT1、TP53、TNF、IL6、VEGFA、MMP9、PTGS2等有关。结论:络石藤可能是通过调节AKT1、TP53、TNF、IL6、VEGFA、MMP9、PTGS2等靶点,发挥抑制炎症反应、调节免疫功能及调控细胞凋亡等作用来治疗RA。Objective:Based on the research method of network pharmacology,to explore the possible mechanism of the treatment of Rheumatoid Arthritis(RA).Methods:Taking oral bioavailability(OB)≥30%and drug-likeness(DL)≥0.18 as indicators,the active ingredients and targets of TCM were obtained through the database of Traditional Chinese Medicine Systems Pharmacology and Analysis Platform(TCMSP),and used The uniprot database converts the corresponding target protein into a standard protein gene name.Search the Disgent database for“Rheumatoid arthritis”for RA disease targets.Using the Venn diagram,the common targets of drug ingredients and RA were obtained,and uploaded to Cytoscape 3.9.1 to construct a network diagram of“Terraphyllum-Compound-Target-Rheumatoid Arthritis”,and conduct protein interaction network analysis(PPI network analysis),GO enrichment and KEGG pathway enrichment analysis.Results:9 components and 109 targets of genus terrazolium were obtained,2722 related targets of RA disease,57 common targets of genus genus and RA,GO functional enrichment analysis showed 394 biological processes(BP),41 There are 1 cellular composition(CC)and 70 molecular functions(MF).KEGG pathway enrichment analysis revealed 132 pathways.Results:This study preliminarily revealed that the six effective active ingredients in Qingfengteng,mainly sinomenine,played an anti-RA role,and the key therapeutic targets were related to AKT1,TP53,TNF,IL6,VEGFA,MMP9,PTGS2,etc.related.Conclusion:Tengshiteng may treat RA by regulating AKT1,TP53,TNF,IL6,VEGFA,MMP9,PTGS2 and other targets,inhibiting inflammatory response,regulating immune function and regulating cell apoptosis.
