摘  要:目的:采用高效液相色谱-电喷雾检测器法(HPLC-CAD),建立同时测定八珍益母丸中盐酸水苏碱、盐酸益母草碱和猪苓酸C含量的检测方法。方法:采用Ultimate XB-Phenyl色谱柱(250 mm×4.6 mm,5μm),以乙腈为流动相A,10 mmol/L甲酸铵(甲酸调pH值至4.0)为流动相B,梯度洗脱,流速为1.0 ml/min,CAD雾化器温度为35℃,采集频率为10 Hz,进样量10μl。结果:分离度均符合要求,阴性无干扰;盐酸水苏碱、盐酸益母草碱和猪苓酸C的线性范围分别为1.10~550.11、0.901~450.26、0.024~12.21μg/ml(r≥0.9991);盐酸水苏碱、盐酸益母草碱和猪苓酸C平均加样回收率分别为99.39%(RSD为0.98%)、98.84%(RSD为0.94%)、100.11(RSD为1.09%),平均加样回收率为98%~102%,RSD≤2.0%。结论:该方法具有良好的灵敏度及准确度,为八珍益母丸的全面质量控制提供科学依据。Objective:To develop an HPLC-CAD method for simultaneous determination of stachydrine hydrochloride,leonurine hydrochloride,and polyporenic acid C in Bazhenyimu pills.Methods:The chromatographic separation was performed on an Ultimate XB-Phenyl column(250 mm×4.6 mm,5μm)with acetonitrile(A)-10 mmol/L ammonium acetate(formic acid tone pH 4.0)(B)as the mobile phase for gradient elution,the flow rate was 1.0 ml/min,the nebulization temperature of the detector was 35℃,the acquisition frequency was 10 Hz,and injection volume was 10μl.Results:The separation of each component was good,negative without interference.The linear ranges of stachydrine hydrochloride,le onurine hydrochloride,and polyporenic acid C were 1.10~550.11,0.901~450.26,and 0.024~12.21μg/ml(r≥0.9991),respectively.The average recoveries of them were 99.39%(RSD of 0.98%),98.84%(RSD of 0.94%),and 100.11(RSD of 1.09%),respectively.The average recoveries were 98%~102%,and the RSD≤2.0%.Conclusion:The method has good sensitivity and accuracy and provides a scientific basis for the overall quality control of Bazhenyimu pills.
