摘 要:对一株野生栓孔菌属的真菌子实体进行分离鉴定,探究其生长特性,并比较菌丝体胞内多糖和发酵液胞外多糖的抗氧化作用、抑癌活性及多糖组分。研究结果显示,该菌株经鉴定为血红栓孔菌Trametes sanguinea,菌丝生长最适条件碳源为甘露糖,氮源为酵母浸提物,pH值为6.0,温度为35℃;胞外多糖比胞内多糖更具有化学抗氧化能力,当多糖浓度为5 mg/mL时,胞内多糖对DPPH、ABTS和羟自由基的清除率分别为57.54%、97.20%和36.40%,铁离子还原力的FRAP值为1.14 mmol/L,而胞外多糖对DPPH、ABTS和羟自由基的清除率分别为63.88%、99.36%和40.99%,FRAP值为2.34 mmol/L;胞外多糖的抑癌活性强于胞内多糖,在供试浓度下多糖溶液对LO2细胞无毒副作用,但能明显抑制Huh7细胞增殖,胞内和胞外多糖的IC50值分别是4.154和2.894 mg/mL;进一步对比分析其单糖的组分和含量,发现除了葡萄糖、盐酸氨基葡萄糖、甘露糖和半乳糖外,胞外多糖比胞内多糖还多了岩藻糖、葡萄糖醛酸和木糖,胞内多糖中仅有葡萄糖含量高于胞外多糖,这表明胞内多糖与胞外多糖活性可能与其单糖组成和含量有关。这为血红栓孔菌资源的进一步开发利用提供参考资料。A wild Trametes strain was isolated and identified.Its growth characteristics and the antioxidative effects and antitumor activities of intracellular polysaccharide in mycelium and extracellular polysaccharides in fermentation broth were investigated.The strain was identified as Trametes sanguinea.The optimum carbon source was mannose,and the nitrogen source was yeast extract.The mycelia grew well at pH value of 6.0 and temperature of 35℃.Extracellular polysaccharide showed more antioxidant capacity than intracellular polysaccharide.Under polysaccharide concentration of 5mg/mL,the scavenging rates of intracellular polysaccharide on DPPH,ABTS and hydroxyl radicals were 57.54%,97.20%and 36.40%respectively,and FRAP value of iron ion reducing power was 1.14 mmol/L,while the scavenging rates of extracellular polysaccharide on DPPH,ABTS and hydroxyl radicals were 63.88%,99.36%and 40.99%respectively,and the FRAP value was 2.34 mmol/L.The antitumor activity of extracellular polysaccharides was stronger than that of intracellular polysaccharides.The polysaccharide solution had no side effects on LO2 cells,but could significantly inhibit the proliferation of Huh7 cells.The IC50 values of intracellular and extracellular polysaccharides were 4.154 and 2.894 mg/mL,respectively.Further analysis of the monosaccharide composition and content found that besides glucose,glucosamine hydrochloride,mannose and galactose,extracellular polysaccharide additionally contained fucose,glucuronic acid and xylose,which were absent in intracellular polysaccharides.In intracellular polysaccharides,only glucose level was higher in comparison with the content of monosaccharide in the extracellular polysaccharide,suggesting that the intracellular and extracellular polysaccharide activity may be related to monosaccharide composition and content.This provides reference for further development and utilization of T.sanguinea resources.
