黄熟胶囊对Ⅱ型糖尿病模型大鼠的影响

摘　　要：目的探讨黄熟胶囊对Ⅱ糖尿病模型大鼠治疗的作用机制。方法连续给予高糖、高脂饲料8周后,再腹腔注射2%链脲佐菌素(STZ),建立Ⅱ型糖尿病大鼠模型。选择模型大鼠,按血糖水平分为模型组、阳性组、黄熟胶囊剂量组。正常组和模型组给予生理盐水,各给药组分别给予不同的药物和剂量治疗8周,给药后检测血糖、胰岛素、甘油三酯和胆固醇。结果正常组大鼠的外观正常,模型大鼠进食与水量和排尿增多、体重下降、毛色枯黄,精神委靡、形体消瘦等特征,治疗后,阳性组及黄熟胶囊组均有好转;模型大鼠经高糖、高脂饲料喂养后,血糖、胰岛素水平极显著地升高,注射STZ后,血糖进一步升高,而胰岛素则降至正常水平;治疗后,模型大鼠的甘油三脂、胆固醇及血糖水平较正常组大鼠极显著升高,胰岛素水平显著低于正常组;阳性组的甘油三脂和胆固醇水平保持高位,血糖较模型组极显著地降低;黄熟胶囊高剂量组血糖、甘油三脂及胆固醇水平较模型组极显著地下降,胰岛素水平显著高于模型组。中剂量组血糖、甘油三脂水平较模型组极显著下降,胆固醇水平较模型组显著下降;低剂量组血糖、甘油三脂显著下降。结论黄熟胶囊在降低高糖、高脂饲料喂养、STZ诱导的胰岛素抵抗模型大鼠血糖、血脂水平的同时对胰岛细胞还有一定的修复作用。这可能是对糖尿病发挥治疗作用的机制之一。OBJECTIVE To investigate the therapeutic mechanism of Huangshu capsules on typeⅡdiabetes model rats.METHODS After 8 weeks of continuous high-glucose and high-fat diet,2%STZ was injected intraperitoneally,and to establish a rat model of type II diabetes.The rats were divided into model group,positive group and Huangshu capsules(high,medium and low dose)groups according to blood glucose level.Normal and model groups were given normal saline,and each administration group was given different drugs and doses respectively for 8 weeks.Blood glucose,insulin,triglyceride and cholesterol were detected after administration.RESULTS The appearance of rats in normal group was normal.The model rats had the characteristics of increased food intake,water volume and urination,decreased body weight,withered and yellow hair color,depressed spirit and emaciated body.After treatment,both the positive group and Huangshu capsules group improved.After the model rats were fed with high-glucose and high-fat diet,the levels of blood glucose and insulin were significantly increased,and the blood glucose was further increased after STZ injection,while the insulin was reduced to normal level.After treatment,triglyceride,cholesterol and blood glucose levels of model rats were significantly higher than those of normal group,and insulin level was significantly lower than that of normal group.Triglyceride and cholesterol levels in the positive group remained high,and blood glucose was significantly lower than that in the model group.The blood glucose and triglyceride levels in the medium-dose group were significantly lower than those in the model group,while the cholesterol level was significantly lower than that in the model group.Blood glucose and triglyceride decreased significantly in low dose group.CONCLUSION Huangshu capsules can reduce blood glucose and blood lipid levels in STZ-induced insulin resistance model rats fed with high-glucose and high-fat diets,and also has certain repair effects on islet cells.This may be one of the mechanism
