摘 要:目的 应用生物信息学方法,分析新冠病毒奥密克戎阳性标本刺突蛋白(S)结构与功能,为抗病毒药物筛选和疫苗设计提供依据。方法 基于盐城市1例奥密克戎阳性标本S蛋白序列,采用生物信息学在线软件,预测分析S蛋白氨基酸组成、理化性质、糖基化修饰、二级结构和受体结合域等生物学特性。结果 该株奥密克戎阳性标本S蛋白为稳定的分泌性蛋白,存在跨膜螺旋区域、具有潜在的186个磷酸化位点、17个糖基化修饰位点和34个抗原细胞表位,蛋白质二级结构以无规则卷曲为主,发现4个有意义的保守结构域。结论 系统性分析奥密克戎毒株S蛋白结构与功能,有助于全面理解S蛋白生物信息学特征。Objective To analyze the structure and protein of the spike(S)protein in a SARS-CoV-2 omicron variant-positive individual using bioinformatics analysis,so as to provide insights into screening of antiviral agents and vaccine design.Methods Based on the sequence of the S protein in an omicron variant-positive sample in Yancheng City,the physiochemical features,glycosyla-tion modification,secondary structure and receptor binding domain were predicted using online bioinformatics tools.Results The S protein in an omicron variant-positive sample from Yancheng City was a stable secretory protein,which had the transmembrane helix re-gion,186 potential phosphorylation sites,17 glycosylation modification sites and 34 antigenic epitopes.The secondary structure of the S protein was characterized by random coil,and four meaningful conserved domains were predicted.Conclusion The structure and pro-tein of the spike(S)protein in a SARS-CoV-2 omicron variant-positive individual were predicted,which facilitates the comprehensive understanding of the bioinformatics features of the S protein.
