LSD1抑制剂联合顺铂对宫颈癌CaSki细胞功能影响的研究

摘　　要：目的探究赖氨酸特异性组蛋白去甲基化酶1(lysine specific demethylase 1,LSD1)特异性抑制剂OGL002与抗癌药物顺铂对宫颈癌CaSki细胞的影响。方法培养CaSki细胞并分为4组:对照组、OGL002组、顺铂组、OGL002+顺铂组,按照组别分别用OGL002以及顺铂处理CaSki细胞48 h。MTT检测细胞增殖,集落形成实验检测细胞增殖,Transwell检测细胞侵袭,流式细胞仪检测细胞凋亡,实时荧光定量PCR检测LSD1的表达,Western blot检测LSD1以及下游组蛋白甲基化水平。结果OGL002与顺铂单独以及联合用药抑制CaSki细胞增殖和侵袭,促进细胞凋亡,降低LSD1的表达,促进H3K4me1、H3K4me2表达水平上调,联合使用效果更加明显。结论OGL002以及顺铂联合用药后降低了LSD1以及组蛋白的表达,并且对细胞功能的影响比单独用药更加明显,提示OGL002与顺铂联合用药对于宫颈癌的治疗具有重要意义。Objective To explore the effects of lysine specific demethylase 1(LSD1)specific inhibitor OGL002 and anticancer drug cisplatin on cervical cancer CaSki cells.Methods CaSki cells were cultured and divided into four groups:control group,OGLO02 inhibitor group,cisplatin group,OGL002+cisplatin group,and CaSki cells were treated with OGL002 and cisplatin for 48 h respectively.MTT assay was used to detect cell proliferation,colony formation assay was used to detect cell proliferation,Transwell assay was used to detected cell invasion,flow cytometry was used to detected apoptosis,real-time fluorescence quantitative PCR was used to detected the expression of LSD1,and Western blot was used to detected LSD1 and downstream histone methylation levels.Results OGLO02 and cisplatin alone and in combination inhibited the proliferation and invasion of CaSki cells,promoted apoptosis,decreased the expression of LSD1,and up-regulated the expression levels of H3K4mel and H3K4me2,and the combined effect was more obvious.Conclusion The combination of OGLO02 and cisplatin reduced the expression of LSD1 and histone,and the effect on cell function was more obvious than that of single drug,suggesting that the combination of OGL002 and cisplatin is of great significance forthetreatmentof cervicalcancer.
