南蛇藤提取物通过PDCD5对异位子宫内膜细胞增殖和凋亡的影响

摘  要:目的 探讨南蛇藤提取物通过程序性细胞死亡因子5(PDCD5)对异位子宫内膜细胞增殖和凋亡的影响。方法 体外分离人异位子宫内膜细胞,采用不同浓度(0、20、40、80μg/mL)的南蛇藤提取物处理,记为南蛇藤提取物各剂量组;按照脂质体法将pcDNA、pcDNA-PDCD5转染细胞内,记为pcDNA组、pcDNA-PDCD5组;将si-NC和si-PDCD5转染至细胞内,然后用80μg/mL的南蛇藤提取物处理,记为南蛇藤提取物+si-NC组和南蛇藤提取物+si-PDCD5组。采用MTT检测细胞增殖;流式细胞术检测细胞凋亡;Western blot检测p21、B淋巴细胞瘤-2(Bcl-2)、bcl-2相关X蛋白(Bax)、PDCD5蛋白表达;RT-qPCR检测PDCD5 mRNA的表达量。结果 南蛇藤提取物各剂量组能降低细胞OD值(48 h值分别为0.063±0.04、0.55±0.05、0.47±0.03、0.38±0.04)、(72 h值分别为0.84±0.07、0.77±0.05、0.65±0.06、0.52±0.04),增加细胞凋亡率(4.2±0.3、8.5±1.1、13.8±1.5、19.7±1.6)%,增加p21(0.22±0.02、0.34±0.04、0.47±0.03、0.62±0.05)、Bax(0.23±0.03、0.36±0.04、0.45±0.04、0.61±0.06)蛋白表达,降低Bcl-2(0.72±0.06、0.65±0.05、0.51±0.04、0.38±0.02)蛋白表达,增加PDCD5 mRNA(0.99±0.07、1.22±0.12、1.51±0.15、1.85±0.16)和蛋白表达(0.32±0.03、0.49±0.05、0.61±0.05、0.72±0.06)。过表达PDCD5降低细胞OD值、Bcl-2蛋白表达,增加细胞凋亡率、p21、Bax蛋白表达,及增加PDCD5 mRNA和蛋白表达。与南蛇藤提取物+si-NC组比较,南蛇藤提取物+si-PDCD5能降低细胞内PDCD5 mRNA和蛋白表达,增加OD值,降低细胞凋亡率,降低p21、Bax蛋白表达,增加Bcl-2蛋白表达。结论 南蛇藤提取物可能通过调控PDCD5抑制人异位子宫内膜细胞增殖和诱导细胞凋亡。ObjectiveeTo investigate the effect of Celastrus orbiculatus extrac on the proliferation and apoptosis of human ectopic endometrial cells through programmed cell death factor 5(PDCD5).Methods Isolate human ectopic endometrial cells in vitro and treat them with different concentrations(0,20,40,80μg/mL)of Celastrus orbiculatus extract,and record them as the different dose groups of Celastrus orbiculatus extract;according to the liposome method,according to the liposome method,pcDNA and pcDNA-PDCD5 were transfected into cells,which were recorded as pcDNA group and pcDNA-PDCD5 group;the si-NC and si-PDCD5 were transfected into the cells,and then treated with 80μg/mL Celastrus orbiculatus extract,which was recorded as Celastrus orbiculatus extract+si-NC group and Celastrus orbiculatus extract+si-PDCD5 group.MTT was used to detect cell proliferation;flow cytometry was used to detect cell apoptosis;Western blot was used to detect p21,Bcl-2,Bax,and PDCD5 protein expression;RT-qPCR was used to detect PDCD5 mRNA expression.Results Each dose group of the Celastrus orbiculatus extract can reduce the cell OD value(the 48 h values were 0.063±0.04,0.55±0.05,0.47±0.03,0.38±0.04),(the 72 h values were 0.84±0.07,0.77±0.05,0.65±0.06,0.52±0.04),increase the apoptosis rate(4.2±0.3,8.5±1.1,13.8±1.5,19.7±1.6)%,increase the expression of p21(0.22±0.02,0.34±0.04,0.47±0.03,0.62±0.05)and Bax(0.23±0.03,0.36±0.04,0.45±0.04,0.61±0.06)protein,decrease the expression of Bcl-2(0.72±0.06,0.65±0.05,0.51±0.04,0.38±0.02)protein,and increase the expression of PDCD5 mRNA(0.99±0.07,1.22±0.12,1.51±0.15,1.85±0.16)andprotein(0.32±0.03,0.49±0.05,0.61±0.05,0.72±0.06).Overexpression of PDCD5 reduces cell OD value,Bcl-2 protein expression,increases cell apoptosis rate,p21,Bax protein expression,and increases PDCD5 mRNA and protein expression.Compared with the Celastrus orbiculatus extract+si-NC group,the Celastrus orbiculatus extract+si-PDCD5 can reduce the expression of PDCD5 mRNA and protein in the cell,increase the OD valu
