摘  要:目的研究白藜芦醇对阿霉素诱导的大鼠肾病综合征的作用和机制。方法将18只雄性SD大鼠随机分为空白组、模型组和实验组。模型组和实验组给予一次性尾静脉注射5 mg·kg^(-1)的阿霉素制备阿霉素肾病大鼠模型。实验组给予20 mg·kg^(-1)白藜芦醇,每天灌胃1次;空白组和模型组均给予等量0.9%NaCl,每天灌胃1次。治疗8周后,用实时荧光定量聚合酶链反应检测肾组织晚期糖基化终末产物受体(AGER)和松弛素(Relaxin)的表达水平,并检测血清肌酸酐和尿素氮等生化指标。结果给予尾静脉注射阿霉素后24 h尿蛋白定量均升高3倍以上,表示造模成功。空白组、模型组和实验组的AGER mRNA相对表达量分别为1.00±0.00、3.30±0.44和2.65±0.17,Relaxin mRNA相对表达量分别为1.00±0.00、0.59±0.07和2.97±0.29,肌酸酐分别为(37.86±5.94)、(39.40±4.47)和(25.95±8.47)μmol·L^(-1),尿素氮分别为(7.40±0.53)、(6.87±0.95)和(7.10±1.01)mmol·L^(-1);实验组的上述指标与模型组相比,差异均有统计学意义(均P<0.05)。结论白藜芦醇通过调控AGER和Relaxin的表达,从而减轻阿霉素诱导的肾病综合征大鼠的肾损伤。Objective To study the effect and mechanism of resveratrol on adriamycin-induced nephrotic syndrome in rats.Methods Eighteen male SD rats were randomly divided into blank group,model group and experimental group.The model group and experimental group were given a single tail vein injection of 5 mg·kg^(-1) adriamycin to prepare adriamycin nephropathy rat model.The experimental group was given 20 mg·kg^(-1) resveratrol by gavage once a day.Blank group and model group were given the same amount of 0.9%NaCl,once a day.After 8 weeks of treatment,the expression levels of receptor of advanced glycation end products(AGER)and Relaxin in renal tissue were detected by real-time fluorescent quantitative polymerase chain reaction,and biochemical indexes such as serum creatinine and urea nitrogen were detected.Results 24 hours after the administration of adriamycin in the tail vein,the urinary protein content increased more than 3 times,indicating successful modeling.The relative expression levels of AGER mRNA in the blank,model and experimental groups were 1.00±0.00,3.30±0.44 and 2.65±0.17;the relative expression levels of Relaxin mRNA were 1.00±0.00,0.59±0.07 and 2.97±0.29;creatine levels were(37.86±5.94),(39.40±4.47)and(25.95±8.47)μmol·L^(-1);urea nitrogen levels were(7.40±0.53),(6.87±0.95)and(7.10±1.01)mmol·L^(-1),respectively.The above indexes in the experimental group were significantly different from those in the model group(all P<0.05).Conclusion Resveratrol reduces renal injury in rats with adriamycin-induced nephrotic syndrome by regulating the expressions of AGER and Relaxin.
