摘 要:目的研究红藻氨酸(KA)对BV2小胶质细胞的增殖和凋亡的影响及机制。方法用KA诱导的BV2小胶质细胞模型。将BV2细胞分为对照组和KA组。对照组用正常的培养基培养,KA组用含600μmol·L^(-1)KA的培养基培养。以酶联免疫吸附试验检测肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL^(-1)β)和IL-6水平,以蛋白质印迹法检测半胱天冬酶3(Caspase-3)、Caspase-9和p53表达水平,以钙成像实验测定细胞内Ca^(2+)浓度。结果KA组的24 h TNF-α、IL-1β、IL-6的含量分别为(130.60±9.22)、(9.83±1.10)和(73.79±11.26)pg·mL^(-1)。对照组和KA组小胶质细胞中Caspase-3蛋白相对表达水平分别为1.00±0.06和1.22±0.09,Caspase-9蛋白相对表达水平分别为1.00±0.14和1.44±0.12,p53蛋白相对表达水平分别为1.00±0.14和1.34±0.13,Ca^(2+)水平分别为1.18±0.29和5.26±0.47。KA组的上述指标与对照组比较,差异均有统计学意义(均P<0.05)。结论KA可以激活小胶质细胞促进其释放炎症因子,高浓度KA可促进BV2小胶质细胞的凋亡,其机制可能与细胞内钙离子升高有关。Objective To study the effect and mechanism of kainic acid(KA)on the proliferation and apoptosis of microglia.Methods BV2 microglia model induced by KA was used.BV2 cells were divided into control group and experimental group.The control group were added normal medium;the KA group were added medium containing 600μmol·L^(-1) KA.Enzyme linked immunosorbent assay(ELISA)was used to detect the release of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),IL-6 after KA stimulation for different time;the expression levels of apoptotic proteins Caspase-3,Caspase-9 and p53 were detected by Western blot;the changes of intracellular calcium concentration in microglia were detected by cell calcium imaging.Result After 24 hours,the contents of TNF-α,IL-1β,IL-6 in KA group were(130.60±9.22),(9.83±1.10)and(73.79±11.26)pg·mL^(-1),respectively.The expression levels of Caspase-3 protein in control and KA groups were 1.00±0.06 and 1.22±0.09,respectively;the expression levels of Caspase-9 protein were 1.00±0.14 and 1.44±0.12,respectively;the expression levels of p53 protein were 1.00±0.14 and 1.34±0.13,respectively;the Ca^(2+)concentrations were 1.18±0.29 and 5.26±0.47,respectively.Compared with the control group,the above indexes in the KA group were significantly different ( all P < 0. 05) . Conclusion KA can activate microglia,and high concentration of KA can promote theapoptosis of BV2 microglia,which may be related to the increase of intracellular calcium.
