摘  要:目的 研究白头翁皂苷A对慢传输型便秘(STC)大鼠的疗效及其作用机制。方法 将SD大鼠按照随机数字表法分为对照组、模型组和低、中、高剂量实验组,每组10只。除对照组大鼠,其余4组大鼠均灌胃给予复方地芬诺酯构建STC大鼠模型,建模成功后,低、中、高剂量实验组分别灌胃给予100、150、300 mg·kg^(-1)白头翁皂苷A,对照组和模型组均灌胃给予等量的0.9%NaCl,每天1次,并记录给药0、6、10、14 d的大鼠体质量变化,连续给药14 d。记录粪便粒数、湿重、干重、含水量。以苏木精-伊红染色法观察结肠黏膜及肠腺杯状细胞数量变化情况,以蛋白质印迹法检测结肠组织黏蛋白2(MUC2)、酪氨酸激酶受体(c-kit)、干细胞因子(SCF)蛋白水平,以酶联免疫吸附法检测5-羟色胺(5-HT)、血管活性肠肽(VIP)、一氧化氮(NO)水平。结果 对照组、模型组和低、中、高剂量实验组大鼠给药14 d后体质量分别为(234.37±3.29)、(200.62±3.91)、(212.59±2.83)、(223.97±3.27)和(225.91±3.27)g,粪便含水量分别为(32.35±6.64)%、(21.98±7.20)%、(28.90±4.37)%、(29.65±7.29)%和(30.07±6.42)%,杯状细胞数分别为(141.10±10.40)、(51.10±5.30)、(62.10±6.20)、(76.80±6.98)和(126.50±7.30)个,MUC2蛋白相对表达水平分别为0.97±0.09、0.32±0.04、0.45±0.04、0.75±0.05和0.95±0.12,5-HT含量分别为(5.14±0.54)、(3.01±0.17)、(4.13±0.41)、(4.50±0.44)和(5.13±0.50)ng·mL^(-1),c-kit相对蛋白水平分别为0.87±0.08、0.31±0.04、0.49±0.07、0.68±0.06和0.08±0.09,SCF相对蛋白水平分别为1.07±0.11、0.38±0.04、0.55±0.05、0.66±0.08和0.89±0.08。模型组的上述指标与对照组比较,低、中、高剂量实验组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。结论 白头翁皂苷A可能通过调控c-kit/SCF通路,调节结肠黏液屏障及肠神经递质水平改善STC大鼠便秘症状。Objective To investigate the efficacy of Pulsatilla saponin A in the treatment of slow transit constipation(STC)rats and its possible mechanism.Methods The SD rats were randomly divided into control group,model group and low,medium,high dose experimental groups,with 10 rats in each group.Except for control group,the rats in other groups were given compound diphenoxylatel by gavage to establish the STC rat model.After successful modeling,the low,medium and high dose experimental groups were given 100,150 and 300 mg·kg^(-1) Pulsatilla saponin A by gavage,respectively.Control group and model group were given the same amount of normal saline once a day,and the body weight changes of rats on 0,6,10,and14 d were recorded. The drug was administered continuously for 14 days. After the last dose,the rats were placed inmetal cages alone to collect feces,and the fecal grain number,wet weight,dry weight and water content wererecorded. Hematoxylin and eosin ( HE) staining was used to observe the number of goblet cells in colonic mucosa andintestinal gland. Western blotting was used to detect the protein levels of mucin 2( MUC2) ,kit proto - oncogene ( c -kit) and stem cell factor ( SCF) in colon tissue. Enzyme - linked immunosorbent assay was used to detect 5 -hydroxytryptamine( 5 - HT) ,vasoactive intestinal peptide( VIP) and nitric oxide( NO) levels. Results After the endof drug administration,the rat body weights in control group,model group and low,medium,high dose experimentalgroups were ( 234. 37 ± 3. 29) ,( 200. 62 ± 3. 91) ,( 212. 59 ± 2. 83) ,( 223. 97 ± 3. 27) ,( 225. 91 ± 3. 27) g;thefecal water content were ( 32. 35 ± 6. 64 ) %,( 21. 98 ± 7. 20 ) %,( 28. 90 ± 4. 37 ) %,( 29. 65 ± 7. 29 ) %,( 30. 07 ± 6. 42) %;the number of goblet cells were 141. 10 ± 10. 40,51. 10 ± 5. 30,62. 10 ± 6. 20,76. 80 ± 6. 98and 126. 50 ± 7. 30;MUC2 relative protein levels were 0. 97 ± 0. 09,0. 32 ± 0. 04,0. 45 ± 0. 04,0. 75 ± 0. 05,0. 95 ± 0. 12;the contents of 5 - HT were ( 5. 14 ± 0. 54 ) ,( 3. 01 ± 0. 17 ) ,( 4.
