摘 要:目的探讨人胎盘间充质干细胞对大鼠子宫内膜损伤的修复作用及对Janus激酶2(JAK2)/信号传导转录激活因子3(STAT3)通路的影响。方法随机将30只8周龄SPF级雌性SD大鼠分为假手术组、模型组、人胎盘间充质干细胞组(以下简称为干细胞组),每组10只。假手术组切开大鼠皮肤,暴露大鼠腹腔后直接缝合切口。模型组与干细胞组大鼠构建子宫内膜损伤模型,随后,干细胞组向宫腔内移植人胎盘间充质干细胞凝胶0.1 ml,模型组不做处理。按雌雄比3∶1进行大鼠分笼。实验结束后,观察各组受孕大鼠子宫内胚胎数,HE染色观察各组大鼠子宫内膜的病理改变,酶联免疫吸附测定法检测各组大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)表达水平,采用实时荧光定量聚合酶链式反应(RT-qPCR)检测各组大鼠子宫内膜组织JAK2、STAT3 mRNA的表达水平,检测各组大鼠子宫内膜组织JAK2、STAT3蛋白水平。结果与假手术组比较,模型组和干细胞组受孕大鼠子宫内胚胎数减少(P<0.05);与模型组比较,干细胞组受孕大鼠子宫内胚胎数增加(P<0.05)。与假手术组比较,模型组大鼠子宫内膜厚度变薄,炎性细胞浸润增加,细胞排列紊乱,同时子宫内膜间质纤维化明显;与模型组比较,干细胞组大鼠子宫内膜病变有明显改善。与假手术组比较,模型组和干细胞组大鼠子宫内膜组织JAK2、STAT3 mRNA及蛋白表达量均减少(P均<0.05),血清TNF-α、IL-1β和IL-6水平均升高(P均<0.05);与模型组比较,干细胞组大鼠子宫内膜组织JAK2、STAT3 mRNA及蛋白表达量增加(P<0.05),血清TNF-α、IL-1β和IL-6水平降低(P<0.05)。结论人胎盘间充质干细胞可有效修复大鼠子宫内膜损伤,其机制可能与人胎盘间充质干细胞激活JAK2/STAT3信号通路,从而降低炎症反应有关。Objective To investigate the effect of human placental mesenchymal stem cells on repairing endometrial injury in rats and its influence on Janus kinase 2(JAK2)/signal transduction activator of transcription 3(STAT3)pathway.Methods Thirty 8-week old specific pathogen free female Sprague Dawley rats were randomly divided into sham operation group,model group,and human placental mesenchymal stem cell group(referred to as stem cell group),with 10 rats in each group.In the sham operation group,the skin of rats was cut,the abdominal cavity of rats was exposed,and the wound were directly sutured.The rats in the model group and the stem cell group were used to construct endometrial injury model,then 0.1 ml human placental mesenchymal stem cell gel was transplanted into the uterine cavity in the stem cell group while the model group were left untreated.The rats were divided into cages according to male to femal ratio of 3∶1.After the experiment,the number of embryos in the uterus of the pregnant rats in all groups was observed.The pathological changes of the endometrium of the rats in all groups were measured by hematoxylin-eosin staining.The expression levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)in the serum of the rats in all groups were detected by enzyme-linked immunosorbent assay.Real time fluorescent quantitative polymerase chain reaction(RT-qPCR)was used to detect JAK2 and STAT3 mRNA in endometrial tissue of rats in all groups,and the levels of JAK2 and STAT3 protein in endometrial tissue of rats in all groups were detected.Results Compared with the sham operation group,the number of embryos in the uterus of the pregnant rats in the model group and the stem cell group were decreased(P<0.05).The number of embryos in the uterus of the pregnant rats in the stem cell group was increased,compared with the model group(P<0.05).Compared with the sham operation group,the endometrial thickness of the rats in the model group was thinner,the infiltration of inflammatory cells was
