摘 要:以豁眼鹅为研究对象,采用荧光定量PCR方法研究Smad1基因mRNA在鹅胚胎期13,18,28 d(E13、E18和E28)3个时间点表达趋势的变化;利用苏木精-伊红染色法(HE染色,Hematoxylin-eosin staining)结合免疫组织化学染色(IHC染色,Immunohistochemistry staining)对皮肤组织进行常规及特异性染色观察。荧光定量PCR结果显示,Smad1基因mRNA相对表达量在E13、E18、E28这3个时间点呈现下降趋势,其中E13与E18相对表达量差异显著(P<0.05),E13与E28相对表达量差异极显著(P<0.01),E13皮肤组织中Smad1基因mRNA相对表达量约为E28组织中表达量的3倍。HE染色和IHC染色结果显示,在E13的组织中,SMAD1主要分布于整个皮肤组织中,包括未分化的真皮与表皮以及真皮凝集体;至E18,SMAD1仍可见于皮肤及毛囊组织,但在毛囊底部呈现阴性状态;E28时,SMAD1阳性表达主要集中于表皮及毛囊周围,其他部位表达明显减少。结果表明:Smad1在鹅胚胎期毛囊发育分化过程中具有潜在的调控作用。Taking Huoyan goose as the research object,the fluorescence quantitative PCR method was used to study the expression trend of Smad1 gene mRNA on the 13th day(E13),18th day(E18)and 28th day(E28)of the embryonic stage of the goose;Hematoxylin-eosin staining(HE staining,Hematoxylin-eosinstaining),combined with immunohistochemical staining(IHC staining,Immunohis⁃tochemistry staining)was used for routine and specific staining of skin tissue.The results of fluores⁃cence quantitative PCR showed that the relative expression of Smad1 gene mRNA showed a down⁃ward trend at the three time points of E13,E18,and E28.The relative expression of E13 and E18 was significantly different(P<0.05),and the relative expression of E13 and E28 is extremely signifi⁃cant(P<0.01).The relative expression of Smad1 gene mRNA in E13 skin tissue is about 3 times that of E28 tissue.The results of HE staining and IHC staining showed that SMAD1 was mainly distrib⁃uted in the whole skin tissues on the 13th day of embryonic stage,including undifferentiated dermis and epidermis,and dermal aggregates;until the 18th day of embryonic stage,SMAD1 was still found in the skin and hair follicle tissues,but showed a negative state at the bottom of the hair follicle.On the 28th day of embryonic stage,the positive expression of SMAD1 was mainly concentrated in the epidermis and hair follicles,and the expression in other parts was significantly reduced.The above ex⁃periments show that SMAD1 has a potential regulatory role in the development and differentiation of goose embryonic hair follicles.
