摘 要:目的为了解南京市2021年蚊虫种类及携带蚊媒病毒感染情况,为蚊媒传染病的预防控制提供基础数据。方法收集南京市2021年2个区蚊虫样本,采用实时荧光定量RT-PCR方法筛查多种蚊媒病毒,采用普通PCR扩增蚊媒病毒的靶基因,从GenBank数据库下载蚊媒病毒各基因型参考株的序列资料,使用Mega 5和BioEdit等生物分析软件分析结果。结果2个区采集三带喙库蚊和白纹伊蚊共5100只,分170次研磨和检测,荧光定量RT-PCR仅在1批次三带喙库蚊检出乙型脑炎病毒,并未在蚊虫检测出其他蚊媒病毒;扩增乙型脑炎病毒PreM/C靶基因,通过核苷酸的亲缘性关系树及核苷酸与氨基酸同源性分析发现为乙型脑炎病毒GI型。结论南京市2021年首次在蚊媒监测到乙型脑炎病毒,相关部门在制定该地区虫媒病毒传播及蚊虫自然感染乙型脑炎病毒防控策略时应考虑上述因素。Objective To understand mosquito species and mosquito-borne virus infection in Nanjing in 2021,and to provide basic data for the prevention and control of mosquito-borne infectious diseases.Methods Mosquito samples were collected from 2 districts in Nanjing in 2021.A variety of various mosquito-borne viruses were detected by real-time fluorescence quantitative PCR,and the target genes of mosquito-borne viruses were amplified by ordinary PCR.The sequence data of the reference strains of each genotype of mosquito-borne virus were downloaded from the GenBank database,and the results were analyzed using bioanalysis software such as Mega 5 and BioEdit.Results A total of 5100 Culex tritaeniorhynchus and Aedes albopictus were collected from 2 districts,ground and tested in 170 batches.Only one strain of Japanese encephalitis virus was detected in Culex tritaeniorhynchus by real-time quantitative PCR,and no other mosquito-borne viruses were detected in the mosquitoes.The target gene PreM/C of Japanese encephalitis virus was amplified and found to be Japanese encephalitis virus type GI by the phylogenetic tree of nucleotide and amino acid homology analysis.Conclusion Japanese encephalitis virus is detected in mosquito vectors for the first time in Nanjing in 2021.The above factors should be taken into account by the relevant authorities when developing strategies for the prevention and control of arbovirus transmission and mosquito vectors naturally infected with Japanese encephalitis virus in the region.
