摘 要:目的观察大黄素对盲肠结扎穿孔小鼠细胞CD4^(+)、CD8^(+)T细胞比率的影响。方法雌性小鼠共48只,按随机数字表法分为正常组、假手术组、盲肠结扎组、大黄素组。各组在造模前72h,灌胃大黄素溶液或0.9%氯化钠溶液,术后6h、12h检测小鼠外周血中性粒细胞计数、淋巴细胞计数、CD4^(+)、CD8^(+)T细胞比率。结果不同组小鼠与正常组比较:假手术组小鼠术后6h、12h中性粒细胞计数、淋巴细胞计数、中性粒细胞与淋巴细胞比值(neutrolphil to lymphocyte ratio,NLR)、CD4^(+)T细胞比率、CD8^(+)T细胞比率、CD4^(+)/CD8^(+)比值差异均无统计学意义(P>0.05);盲肠结扎组小鼠术后6h中性粒细胞计数(P=0.008)、NLR(P=0.002)、CD8^(+)T细胞比率(P=0.014)显著升高,淋巴细胞计数(P<0.001)、CD4^(+)T细胞比率(P=0.008)和CD4^(+)/CD8^(+)比值(P=0.002)显著降低;大黄素组小鼠术后6h中性粒细胞计数(P<0.001)、NLR(P=0.004)、CD8^(+)T细胞比率(P=0.04)显著升高,淋巴细胞计数(P=0.026)显著降低,CD4^(+)T细胞比率(P=0.134)、CD4^(+)/CD8^(+)比值(P=0.083)无明显变化;盲肠结扎组小鼠术后12h中性粒细胞计数(P<0.001)、NLR(P<0.001)和CD8^(+)T细胞比率(P<0.001)显著升高,淋巴细胞计数(P=0.001)、CD4^(+)T细胞比率(P<0.001)和CD4^(+)/CD8^(+)比值(P<0.001)显著降低;大黄素组小鼠术后12h中性粒细胞计数(P<0.001)、NLR(P=0.001)显著升高,淋巴细胞计数(P=0.028)、CD4^(+)T细胞比率(P=0.005)和CD4^(+)/CD8^(+)比值(P=0.012)显著降低,CD8^(+)T细胞比率无变化(P=0.132)。大黄组小鼠与盲肠结扎组比较:大黄素组小鼠术后6h中性粒细胞计数(P=0.003)、CD4^(+)T细胞比率(P=0.001)、CD4^(+)/CD8^(+)比值(P=0.001)显著升高,淋巴细胞计数(P=0.003)和CD8^(+)T细胞比率(P=0.021)显著降低,NLR(P=0.248)无明显变化;大黄素组小鼠术后12h中性粒细胞计数(P=0.014)、NLR(P=0.015)、CD8^(+)T细胞比率(P=0.001)显著升高,淋巴细胞计数(P=0.021)、CD4^(+)T细胞比率(P=0.005)Objective To observethe effect of emodin on the CD4^(+)and CD8^(+)T cells in mice aftercecal ligation puncture(CLP).Methods A total of 48 female mice were divided into normal group,sham operation group,CLP group,and emodin group according to the random number table method.Intragastric administration of emodin solution or 0.9%sodium chloride solution in various groups of mice 72 hours before the operation.The proportion of neutrophil,lymphocyte,CD4^(+),and CD8^(+)T cells of mice was tested at 6 hours and 12 hours after the operation.Results Compared with the normal group at 6 hours and 12 hours,there were no significant differences in the neutrophil count,lymphocytes,neutrophil-to-lymphocyte ratio(NLR),CD4^(+)T cell ratio,CD8^(+)T cells,and CD4^(+)/CD8^(+)in sham operation group at 6 hours and 12 hours(P>0.05).Compared with the normal group at six hours,the neutrophils(P=0.008),NLR(P=0.002),CD8^(+)T cell ratio(P=0.014)in the CLP groupsignificantly increased at 6 hours after the operation,and the lymphocytes(P=0.014),CD4^(+)T cell ratio(P=0.008),and CD4^(+)/CD8^(+)ratio(P=0.002)significantly decreased.Neutrophils(P<0.001),NLR(P=0.004),and CD8^(+)T cell ratio(P=0.04)in the emodin groupsignificantly increased at 6 hours,lymphocytes(P=0.026)significantly decreased,with CD4^(+)T cell ratio(P=0.134),CD4^(+)/CD8^(+)ratio(P=0.083)had no significant change.Compared with the normal group at 12 hours,the neutrophils(P<0.001),NLR(P<0.001),and CD8^(+)T cell ratio(P<0.001)in the CLP group significantly increased,the lymphocytes(P<0.001),CD4^(+)T cell ratio(P<0.001),and CD4^(+)/CD8^(+)ratio(P<0.001)significantly decreased.Neutrophils(P<0.001),NLR(P=0.001)in the emodin group significantly increased,lymphocytes(P=0.028),CD4^(+)T cell ratio(P=0.005),and CD4^(+)/CD8^(+)ratio(P=0.012)significantly decreased at 12 hours,and there was no change in CD8^(+)T cell ratio(P=0.132).Compared with the CLP group at 6 hours after the operation,the ratio of neutrophils(P=0.003),CD4^(+)T cells(P=0.001),and CD4^(+)/CD8^(+)ratio(P=0.001)of mice in t
