摘 要:旨在探究丹皮酚对脂多糖(lipopolysaccharide,LPS)诱导的奶牛乳腺上皮细胞(bovine mammary epithelial cells,BMECs)炎性损伤的缓解作用。使用CCK-8法检测不同浓度丹皮酚对BMECs活力的影响,选用20、50和100 mg/L丹皮酚处理LPS诱导的BMECs炎性模型,并检测细胞凋亡、氧化应激因子、相关炎性因子及NF-κB关键通路蛋白表达的变化。结果显示:10~100 mg/L丹皮酚对BMECs增殖有不同程度的促进作用;不同剂量的丹皮酚可显著抑制细胞凋亡及IL-6、IL-1β、TNF-α的mRNA表达水平,提高超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性,降低丙二醛(MDA)含量,抑制IκBα和p65的磷酸化水平。以上结果表明,丹皮酚能够抑制LPS诱导的BMECs的炎性损伤,其作用机制可能与NF-κB信号通路被抑制有关。This study was to determine the alleviating effect of Paeonol on the inflammatory injury induced by lipopolysaccharide(LPS)in bovine mammary epithelial cells(BMECs).The effects of Paeonol on BMECs viability were determined using the CCK-8 method,and the LPS-induced BMECs inflammatory model was treated with 20,50,and 100 mg/L Paeonol,respectively;and the changes in the cell apoptosis,the contents of oxidative stress factors,the expressions of related inflammatory factors and the key proteins in the NF-κB pathway were detected.The results showed that 10-100 mg/L Paeonol stimulated BMECs generation to a different extent.The cell apoptosis,the MDA content,the mRNA expressions of IL-6,IL-1β,TNF-α and the phosphorylation levels of IκBαand p65 reduced significantly at different dosages of Paeonol.Meanwhile,the activities of SOD and GSH-Px increased significantly in the above dosages.The present results suggested that Paeonol might inhibit LPS-induced pro-inflammatory injury by suppressing the NF-κB signaling pathways in BMECs.
