摘  要:目的:观察黄芪甲苷(AS-Ⅳ)在葡聚糖硫酸钠(DSS)诱导的小鼠溃疡性结肠炎(UC)中的保护作用并初步探究其保护机制。方法:将雄性C57BL/6小鼠随机分为Control组和AS-Ⅳ组,AS-Ⅳ组连续灌胃给予AS-Ⅳ(100 mg/kg)7 d。证实AS-Ⅳ对小鼠体质量和结肠长度均无影响后,将雄性C57BL/6小鼠随机分为Control组、DSS组、DSS+AS-Ⅳ组,DSS组小鼠自由饮用2.5%DSS水溶液7 d,DSS+AS-Ⅳ组小鼠自由饮用2.5%DSS水溶液的同时连续灌胃给予AS-Ⅳ(100 mg/kg)7 d。HE染色观察小鼠结肠组织病理变化;qRT-PCR检测小鼠结肠组织细胞因子mRNA表达;流式细胞术检测小鼠外周血和结肠固有层单核/巨噬细胞比例。结果:与Control组相比,AS-Ⅳ组小鼠体质量和结肠无明显变化,DSS组小鼠体质量明显减轻、结肠长度缩短,肠黏膜组织结构破坏,炎症细胞浸润增多,结肠组织TNF-α、IL-1β、IL-6、CCL2、CCR2 mRNA表达显著升高;与DSS组相比,DSS+AS-Ⅳ组小鼠结肠缩短程度和结肠组织炎症细胞浸润水平降低,Ly6C+MHCⅡ-巨噬细胞和外周血炎症性单核细胞比例降低,结肠组织TNF-α、IL-1β、IL-6、CCL2、CCR2 mRNA表达显著降低,而结肠组织IL-10、TGF-β mRNA表达显著升高。结论:AS-Ⅳ通过降低DSS诱导的UC小鼠结肠固有层中Ly6C+MHCⅡ-巨噬细胞比例及炎症因子表达减缓了UC发生发展。Objective:To investigate protective effects of AstragalosideⅣ(AS-Ⅳ)in mice with ulcerative colitis(UC)induced by dextran sulfate sodium(DSS)and to preliminarily explore its protective mechanisms.Methods:Male C57BL/6J mice were randomly classified into Control group and AS-Ⅳgroup,AS-Ⅳgroup was continuously given AS-Ⅳ(100 mg/kg)for 7 days through gavage.After marginal effects of AS-Ⅳon mice body weight and colon length has been verified,male C57BL/6 mice were further ran‐domly divided into Control group,DSS group,and DSS+AS-Ⅳgroup.Mice in DSS group freely drank 2.5%DSS aqueous solution for 7 days while mice in DSS+AS-Ⅳgroup were continuously gavaged with AS-Ⅳ(100 mg/kg)based on freely drank 2.5%DSS aqueous solution for 7 days.HE staining was used to observe pathological changes of colon structure of mice;qRT-PCR was used to detect mRNA expressions of cytokines in mice colon tissues;flow cytometry was used to detecte ratio of monocytes/macrophages in peripheral blood and colonic lamina propria of mice.Results:Compared with Control group,body weight and colon of mice in AS-Ⅳgroup had no obvious changes,while body weight of mice in DSS group was decreased dramatically and length of colon was shorter,intestinal mucosal tissue structures were destroyed,inflammatory cell infiltration was increased,and mRNA expressions of TNF-α,IL-1β,IL-6,CCL2 and CCR2 in colon tissue were significantly elevated.Compared with DSS group,degree of colon shortening and level of inflam‐matory cell infiltration in colon tissue of mice in DSS+AS-Ⅳgroup were decreased,proportion of Ly6C+MHCⅡ−macrophages and inflammatory mononuclear cells in peripheral blood were decreased.mRNA expressions of TNF-α,IL-1β,IL-6,CCL2 and CCR2 were significantly decreased,while mRNA levels of IL-10 and TGF-βin colon tissue were significantly increased.Conclusion:AS-Ⅳslows progress and development of UC by decreasing proportion of Ly6C+MHCⅡ−macrophages in colonic lamina propria and expressions of inflammatory cytokines in mice
