摘 要:为了研究幽门结扎和乙醇诱导两种胃溃疡模型对酸浆乙酸乙酯活性部位抗胃溃疡的作用机理,采用幽门结扎胃溃疡模型测定胃液量、游离酸、总酸量及蛋白酶活性,采用乙醇诱导胃溃疡模型检测血浆中一氧化氮(NO)、丙二醛(MDA)、超氧化物歧化酶(SOD)活性和组织匀浆液中表皮生长因子(EGF)的表达;通过蛋白质印迹法(Western blot)和实时荧光定量核酸扩增检测系统(QPCR)分析胃粘膜中环氧化酶1和2(COX-1/2)的蛋白表达和mRNA表达。结果显示:EAF以100、250和500 mg/kg 3个剂量灌胃时,均能显著降低胃液量、游离酸、总酸和胃蛋白酶活性(P<0.05)。EAF中、高剂量组能显著增加血浆中NO含量和SOD活性(P<0.05),降低MDA水平(P<0.01),上调胃组织匀浆液中EGF表达(P<0.05)。关于EAF对COX蛋白和mRNA表达的影响,COX-1的表达水平无显著差别(P>0.05),EAF各剂量组与模型组和对照组相一致。但相对于模型组,EAF各剂量组明显抑制COX-2蛋白和mRNA表达的上调(P<0.05)。EAF可能通过对大鼠胃液量、游离酸度、总酸度和胃蛋白酶活性的抑制,促进氧自由基清除剂SOD、保护因子NO和EGF的合成,降低MDA的损害和抑制COX-2信号通路,从而提高溃疡愈合质量。This study was designed to investigate the underlying mechanism of EAF on gastric ulcer induced by ethanol by pyloric ligation and ethanol induced gastric ulcer model.SD rats were selected as the study object,and the gastric juice volume,dissociate acid,the total acidity and stomach proteinase activity were detected by establishing the experimental model of pylorus ligation induced gastric ulcer.In the ethanol-induced acute gastric ulcer experimental model,the contents of nitric oxide(NO),malondialdehyde(MDA)and the activities of the superoxide dismutase(SOD)from plasma and the expression of EGF from the gastric mucosa around the ulcer were all determined.In the meantime,the expression of gastric mucosa COX-1/COX-2 protein and COX-1/COX-2 mRNA was analyzed by Western blot and QPCR.Results showed that EAF could decrease the contents of dissociate acid,the total acidity and stomach proteinase activity at doses of 100,250 and 500 mg per kg,p.o.body weight.In the ethanol-induced acute gastric ulcer model,EAF at the doses of 250 and 500 mg/kg,p.o.significantly increased the contents of NO and the activity of SOD(P<0.05),decreased the level of MDA(P<0.01),and up-regulated the expression of EGF from tissue homogenate(P<0.05).As for the effect of EAF on the expression of COX mRNA and protein,the expression of COX-1 protein and mRNA showed no difference among the three test groups and was identical with the control and the standard groups.However,relative to the model group,EAF inhibited the up-regulation of the expression of COX-2 protein and mRNA(P<0.05).This study suggested that via inhibiting the gastric juice,free acidity,total acidity and pepsin activity in rats,EAF could promote the synthesis of oxygen radical scavenger SOD,protective factor NO and EGF,and inhibit the signaling pathway of COX-2.and thus improving the healing quality of ulcer.
