摘  要:试验旨在研究中国南方荷斯坦牛触珠蛋白(Haptoglobin,HP)基因的多态性及其对产奶性状、血液生理生化指标的影响。利用直接测序法及SNaPshot技术检测HP基因编码区(Coding sequence,CDS)的单核苷酸多态性(Single nucleotide polymorphism,SNP)位点,并对这些多态性位点不同基因型和组合与785头中国南方荷斯坦牛的产奶性状及血液生理生化指标进行关联分析。结果显示,HP基因的CDS区域存在3个SNPs位点,即rs41871650、rs209034260和rs41871651,这3个位点均处于Hardy-Weinberg平衡状态。在试验群体中,rs41871650位点对305 d奶量、总奶量和高峰奶性状的效应达到显著水平(P<0.05);rs209034260位点对总奶量和高峰奶性状的效应达到显著水平(P<0.05)。rs41871650位点对天门冬氨酸转氨酶(AST)影响极显著(P<0.01),对淋巴细胞百分比(LYMPH%)、白蛋白(ALB)、丙氨酸转氨酶(ALT)及碱性磷酸酶(ALP)影响显著(P<0.05);rs41871651位点对LYMPH%和AST影响极显著(P<0.01),对红细胞平均血红蛋白浓度(MCHC)和甘油三酯(TG)影响显著(P<0.05)。HP基因rs41871650及rs209034260位点的优势基因型组合GG-CC和GG-TT个体305 d产奶量和高峰奶显著高于GA-CT组合个体(P<0.05),AST极显著低于AA-TT和GA-TT组合个体(P<0.01),ALP显著低于AA-TT组合个体(P<0.05)。说明HP基因可以作为影响中国荷斯坦牛产奶性状和健康性状的候选基因用于标记辅助选择。This study was aimed to investigate the SNP of Haptoglobin(HP)gene in Holstein cow in Southern China and its effects on milk production traits and hematology and serum biochemical indexes.Direct sequencing and SNaPshot assay were applied to detect the polymorphisms in coding sequence(CDS)region of HP gene.Then association of the polymorphic sites with the milk production traits and hematology and serum biochemical indexes of 785 Holstein cows in Southern China was analyzed.The results showed that three SNPs were identified,including rs41871650,rs209034260 and rs41871651 in CDS region.All of them were in Hardy-Weinberg equilibrium.The rs41871650 SNP had significant effect on 305 d milk yield,total milk yield and peak milk(P<0.05),and the rs209034260 SNP had significant effect on total milk yield and peak milk(P<0.05).The rs41871650 SNP had highly significant effect on AST(P<0.01),and significant effect on LYMPH%,ALB,ALT and ALP(P<0.05).The rs41871651 SNP had highly significant effect on LYMPH%and AST(P<0.01),and significant effect on MCHC and TG(P<0.05).The GG-CC and GG-TT genotype combinations at rs41871650 and rs209034260 SNPs had significantly greater 305 d milk yield and peak milk than those with GA-CT(P<0.05),and extremely lower AST than those with AA-TT and GA-TT(P<0.01),and significantly lower ALP than those with AA-TT(P<0.05).Therefore,HP gene might be useful molecular markers for milk production traits and health traits in Holstein cow breeding in China.
