摘 要:热休克蛋白70(Heat shock protein 70, Hsp70)是一种在进化上高度保守的分子伴侣蛋白,参与蛋白质的合成、折叠、组装及降解等过程,还参与细胞抗逆境胁迫的响应,对细胞蛋白内稳态(Protein homeostasis)的维持和生物体健康具有重要作用。研究表明,Hsp70对病毒的感染和复制也发挥重要作用。本文就Hsp70的结构、Hsp70对病毒复制的促进及抑制作用进行综述,以期更深入地阐明Hsp70发挥作用的分子机制并为寻找以Hsp70为靶点的抗病毒药物提供新的理论依据及思路。Heat shock protein 70(Hsp70) is an evolutionally highly conserved protein that acts as a “molecular chaperone”. It is involved in the synthesis, folding, assembly and degradation of proteins. It is also involved in the response of cells to stress, and plays a significant part in the maintenance of cellular protein homeostasis and the health of organisms. Studies have indicated that Hsp70 also has an important role in the infection and replication of viruses. Here, the structure of Hsp70 as well as the promotion and inhibition effects of Hsp70 on virus replication are reviewed. This review aims to:(i) elucidate the molecular mechanism of action of Hsp70;and(ii) provide a new theoretical basis and ideas for screening antiviral drugs targeting Hsp70.
