摘 要:研究特女贞苷对氧化应激诱导的成骨细胞模型的保护作用及作用机制。试验采用H2O2刺激MC3T3-E1细胞诱导氧化损伤作为细胞模型,不同浓度的特女贞苷(50μM、10μM、1μM)作用于细胞24 h后,分别检测细胞活性、细胞上清液中碱性磷酸酶(ALP)水平以及骨保护素(OPG)和核因子κB受体活化剂配体RANKL蛋白分泌量。结果显示,与H2O2模型组相比,50μM和10μM特女贞苷组的细胞活性显著升高(P<0.05),1μM特女贞苷组的OPG蛋白分泌显著升高(P<0.05)。随后以特女贞苷(50μM)和P38MAPK通路抑制剂(1μM)混合作用于细胞24 h,结果显示,添加抑制剂后细胞活性、ALP水平和OPG蛋白分泌均呈现显著的抑制趋势。结果提示,特女贞苷在一定浓度范围内对氧化应激状态下的成骨细胞具有保护作用,并且其保护作用可能与P38MAPK信号通路相关。To study the protective effect and mechanism of specnuezhenide on osteoblast model induced by oxidative stress.MC3T3-E1 cells were stimulated by H2O2to induce oxidative damage as a cell model.The cell activity,the level of alkaline phosphatase(ALP)in the supernatant and the secretion of OPG and RANKL were detected after the cells were treated with different concentrations of privet privet(50μM,10μM,1μM)for 24 h.The results showed that compared with H2O2model group,the cell activity in 50μM and 10μM specnuezhenide groups was significantly increased(P<0.05)and the OPG protein secretion in 1μM specnuezhenide group was significantly increased(P<0.05).Then the cells were treated with the mixture of specnuezhenide(50μM)and P38MAPK pathway inhibitor(1μM)for 24 h.The results showed that after treated with the inhibitor,the cell activity,ALP level and OPG protein secretion showed a significant inhibitory trend.The results suggest that privet has a protective effect on osteoblasts under oxidative stress in a certain concentration range,and its protective effect may be related to the P38MAPK signaling pathway.
