摘 要:以槟榔品种热研1号为研究对象,分别以成熟合子胚、一年生种苗茎尖和无菌苗幼根为外植体,研究了不同浓度的植物激素对愈伤诱导、体胚诱导、再生芽萌发及生根的影响,最终获得槟榔组培苗。结果表明:在愈伤诱导阶段,2,4-D为120 mg/L时,茎尖、合子胚、幼根的愈伤诱导率均最高,分别为83.34%,53.34%和10.00%;在体胚诱导阶段,6-BA为50 mg/L时,体胚萌发率最高,为73.33%;在再生芽萌发及生根阶段,NAA为1 mg/L时,体胚萌发率和成苗率最高,分别为83.33%和73.33%。本研究初步建立了槟榔组织培养技术体系,为后续槟榔的组培快繁和遗传转化提供技术支持。In this study,the explants for tissue culture were mature zygotic embryos,shoot tips of one-year seedlings,and the young roots of tube seedlings of the cultivar‘Reyan 1’.The effects of different concentrations of hormones on callus induction,somatic embryo induction,regenerated bud germination,and rooting were investigated,and areca nut tissue culture seedlings were obtained through somatic embryogenesis.The results showed that in the callus induction stage,the highest callus induc-tion rate of shoot tips,zygotic embryos,and young roots was obtained in the medium with 120 mg·L^(-1)2,4-D,83.34%,53.34%,10.00%,respectively.In the somatic embryo induction,the somatic embryo induction rate of medium with 50 mg/L 6-BA was the highest(73.33%).In the regenerated bud germination and rooting stage,the highest germination rate(83.33%)and seed-ling rate(73.33%)were found in the medium with 1 mg·L^(-1)NAA.The tissue culture technical system of the areca nut was ini-tially established,which could provide technical support for the rapid propagation and genetic transformation of the areca nut.
