电针对MCAO大鼠缺血脑组织VEGF和COX-2表达的影响及二者相关性研究

摘  要:目的研究电针对大脑中动脉闭塞(MCAO)模型大鼠缺血中心区和半暗带区血管内皮生长因子(VEGF)和环氧化酶-2(COX-2)的表达的影响及二者之间的相关性。方法制备MCAO老鼠模型,予以电针“百会”“水沟”“足三里”进行干预,应用免疫组化法分别于不同时间点观察各组MCAO老鼠缺血脑组织中心区和半暗带区VEGF和COX-2的表达,分析电针对它们的影响,并探索二者之间是否存在一定的相关性。结果电针组在缺血中心区早期VEGF呈强阳性表达,随着时间延长,表达逐渐下降;在缺血半暗带区先逐渐增多,至72h时达峰值,然后逐渐下降,同时相电针组的阳性表达数均较模型组多(P<0.05)。COX-2在缺血中心区随着时间的延长,表达逐渐增多;在缺血半暗带区早期呈强阳性表达,随着时间延长,表达逐渐下降,至72h时表达量最少,随后又增加,同时相电针组阳性细胞表达数均较模型组少(P<0.05)。结论电针百会、水沟和足三里,可促进VEGF表达,抑制COX-2表达,二者在促进新血管生成、修复损伤神经过程中呈负相关性。Objective To study the effect of electroacupuncture on the expression of vascular endothelial growth factor(VECF)and cyclooxygenase-2(COX-2)in ischemic core and penumbra of middle cerebral artery occlusion(MCAO)rats and the relationship between them.Methods Established the MCAO mouse models and electroacupuncture at"Baihui","Shuigou"and"Zusanli".Immunohistochemical method was used to observe the expression of VECF and COX-2 in the central area and penumbra area of ischemic brain tissue of MCAO rats at different time points,to analyze the effect of electroacupuncture on them,and to explore whether there was a certain correlation between them.Results In the electroacupuncture groups,the expression of VEGF were strongly positive in the early stage of the ischemic center,and decreased gradually with the prolongation of time;And gradually increased in the ischemic penumbra,reached the peak at 72 h,and then decreased gradually.At the same time,the number of positive expression in the electroacupuncture groups were more than that in the model groups(P<0.05).The expression of COX-2 in the ischemic center increased gradually with the prolongation of time;The number of positive expression in the ischemic penumbra increased gradually,reached the pea kelectroacupuncture Baihui,Shuigou and Zusanli can promote the expression of VEGF and at 72 h,and then decreased gradually,at the same time,the number of positive expression in the groups were more than that in the model groups(P<0.05).Conclusion Electroacupuncture at inhibit the expression of COX-2.There is a negative correlation between them in the process of promoting angiogenesis and repairing injured nerve.
