摘 要:目的:分析放射性涎腺损伤大鼠经骨髓间充质干细胞修复后的效果。方法:选取30只大鼠,10只作为空白组,剩余20只建立放射性涎腺损伤大鼠模型,将建模大鼠分为放射对照组、实验组各10只。实验组给予100μl骨髓间充质干细胞悬液,腹腔注射,1次/d,共注射7 d,空白组、放射对照组不做干预,正常喂养。检测对比各组大鼠唾液流量、腮腺指数、下颌下腺指数、水通道蛋白5(AQP5)mRNA表达量及蛋白相对表达量。结果:与空白组相比,放射对照组、实验组唾液流量、腮腺指数、下颌下腺指数水平、AQP5 mRNA表达量、蛋白相对表达量下降(均P<0.05);与放射对照组相比,实验组唾液流量、腮腺指数、下颌下腺指数水平、AQP5 mRNA表达量、蛋白相对表达量上升(均P<0.05)。结论:放射性涎腺损伤大鼠经骨髓间充质干细胞干预后唾液流量、腮腺指数、下颌下腺指数上升,其机制可能与AQP5表达量增加有关。Objective:To investigate the effect of bone marrow mesenchymal stem cells(BMSCs)in repairing radiation-induced salivary gland injury in rats.Methods:Thirty rats were selected,10 rats were taken as the blank group,and the remaining 20 rats were used to establish the radiation-induced salivary gland injury rat model.The model rats were divided into the radiation control group and the experimental group,with 10 rats in each group.The experimental group was given 100μl bone marrow mesenchymal stem cell suspension by intraperitoneal injection,once a day for 7 days.The blank group and the radiation control group were not intervened and fed normally.The salivary flow,Parotid Gland Index,Submandibular Gland Index,and the relative expression of aquaporin 5(AQP5)mRNA and protein were detected and compared among the groups.Results:Compared with the blank group,the salivary flow,Parotid Gland Index,Submandibular Gland Index,AQP5 mRNA and protein expression in the radiation control group and the experimental group were decreased(all P<0.05).Compared with the radiation control group,the salivary flow,Parotid Gland Index,Submandibular Gland Index,AQP5 mRNA expression and protein expression in the experimental group were increased(all P<0.05).Conclusion:The salivary flow,Parotid Gland Index and Submandibular Gland Index of rats with radiation-induced salivary gland injury are increased after BMSCs intervention,and the mechanism may be related to the increase of AQP5 expression.
